摘  要:目的通过观察不同温度对人造血干细胞(HSCs)生长及凋亡情况的影响,探讨不同温度对HSCs生长的抑制情况。方法将HSCs在不同温度下培养分为5组:33℃组、35℃组、37℃组、39℃组、41℃组,其中37℃组作为对照组。分别培养4、7、10d,观察红系集落形成单位(CFU-E)集落数;分别培养6、12、24h,观察HSCs细胞凋亡情况。结果培养4d,37℃组CFU-E集落数最高,33℃组和35℃组与37℃组比较差异有统计学意义(P<0.05);培养7d,37℃组CFU-E集落数最高,33℃组、41℃组与37℃组比较差异有统计学意义(P<0.05);培养10d后,37℃组CFU-E集落数最高,33℃组与37℃组比较差异有统计学意义(P<0.05)。培养6h,33℃组HSCs凋亡率最低,33℃组、35℃组与37℃组比较差异有统计学意义(P<0.05);培养12h,33℃组HSCs凋亡率最低,35℃组、39℃组与37℃组比较差异有统计学意义(P<0.05);培养24h,33℃组HSCs凋亡率最低,35℃组、41℃组与37℃组比较差异有统计学意义(P<0.05)。培养6、12、24h,温度变化与流式细胞仪检测HSCs凋亡率均具有相关性(P<0.01)。结论不同温度对HSCs的生长和凋亡有影响。37℃能促进HSCs的生长,温度降低可以抑制HSCs的凋亡。Objective To investigate the inhibiting situation of different temperatures on the growth of human hematopoietic stem cells (HSCs) by observing the influence of different temperatures on growth and apoptosis of HSCs. Methods HSCs cultured under different temperatures were divided into 5 groups:33 ℃, 35 ℃ ,37 ℃,39 ℃ and 41 ℃,in which the 37 ℃ group served as the control group,and cultured for 4,7,10 d.The colony number of the erythroid colony forming unit (CFU-E) was observed and the apoptotic situation of HSCs after culture for 6,12,24 h was observed. Results After 4 d culture,the colony number of CFU-E in the 37 ℃ group was the highest,which had statistical difference between the 33 ℃ group and 35 ℃ group with 37 ℃ group;after 7 d culture,the colony number of CFU-F in the 37 ℃ group was the highest,which had statistical difference between the 33 ℃ group and 41℃ group with the 37 ℃ group ( P 〈0.05);after 10 d culture,the colony number of CFU-E in the 37 ℃ group was the highest,which had statistical difference between the 33 ℃ group and 37 ℃ group ( P 〈0.05).After 6 h culture,the apoptotic rate of HSCs in the 33 ℃ group was the lowest,which had statistical difference between the 33 ℃ group and 35 ℃ group with the 37 ℃ group ( P 〈 0.05 );after 12 h culture,the apoptotic rate of HSCs in the 33 ℃ group was the lowest,which had statistical difference between the 35 ℃ group and 39 ℃ group with the 37 ℃ group ( P 〈0.05);after 24 h culture,the apoptotic rate of HSCs in the 33 ℃ group was the lowest,which had statistical difference between the 35 ℃ group and 41 ℃ group with the 37 ℃ group ( P 〈0.05).After culture for 6,12,24 h,the temperature change had the correlation with HSCs apoptosis rate detected by flow cytometry. Conclusion Different temperatures have the effect on the growth and apoptosis of HSCs.37 ℃ can promote the growth of HSCs and the temperature decrease can inhibit the apoptosis of HSCs.
