摘  要:目的研究一点红对链脲佐菌素(STZ)致2型糖尿病大鼠肾脏的保护作用及机制。方法 SD雄性大鼠高脂高糖饲料喂养2周后,腹腔注射STZ 35 mg/kg,建立2型糖尿病模型大鼠。将模型大鼠随机分为模型组、厄贝沙坦(0.015 8 g/kg)组及一点红提取物(10 g/kg)组,连续灌胃给药4周。于实验结束前1 d,记录各组大鼠24 h的饮食量、饮水量及尿量;测定各组大鼠尿液中24 h尿蛋白总量(UPro)、24 h尿白蛋白总量(UAlb)、尿肌酐(Ucr)及尿液渗透压(UOP)等;测定血液中尿素氮(BUN)、血肌酐(Scr)、尿酸(UA)、白蛋白(Alb)、三酰甘油(TG)及动脉收缩血压(SBP)等;取双肾,计算各组大鼠的肾脏肥大指数(KI);观察各组大鼠的肾脏病理切片;免疫组化测定各组大鼠肾组织中转化生长因子β1(TGF-β1)和骨形态发生蛋白(BMP-7)的表达。结果模型组大鼠的24 h饮食量、饮水量、尿量、UPro、UAlb、Ucr、UOP、BUN、Scr、UA、TG、SBP、肾重、KI以及TGF-β1、BMP-7表达与正常对照组比较差异有统计学意义(P <0.05),病理改变明显;治疗后,厄贝沙坦组、一点红提取物组大鼠的24h饮食量、饮水量、尿量、UPro、UAlb、Ucr、SUN、Scr、UA、TG、SBP、肾重、KI及TGF-β1表达低于模型组(P <0.05),UOP、BMP-7表达高于模型组(P <0.05),且肾脏病理改变较模型组改善;除Ucr、TG外,厄贝沙坦组和一点红提取物组之间的各项指标比较差异均无统计学意义(P> 0.05)。结论一点红可改善2型糖尿病大鼠肾脏病变,对肾脏有一定的保护作用,其机制可能与调控糖尿病肾组织的TGF-β1、BMP-7的表达失衡有关。Objective To study the protective effect and its mechanism of Emilia sonchifolia on kidneys of rats with type 2 diabetes mellitus induced by streptozotocin. Methods Male SD rats were intragastrically(ig) administered with high-fat and high-sugar diet for 2 weeks,and the type 2 diabetic model was established by intraperitoneal injection of streptozotocin(STZ) 35 mg/kg. Rats with type 2 diabetes mellitus were randomly divided into model group, irbesartan group( 0. 015 8 g/kg) and Ernilia sonchifolia group (10 g/kg ) , and all animals were ig administered once daily for 4 weeks. On the day before the end of the experiment,the 24 h food intake,water intake and urine volume in rats of each group were recorded. The total 24 h urinary protein ( UPro ) , 24 h urinary albumin ( UAlb ) , urinary creatinine(Ucr) and urine osmotic pressure(UOP) were measured. Blood urea nitrogen(BUN) , serum creatinine(Scr) ,uric acid (UA) , albumin ( Alb ) , triglyceride ( TG ) and systolic blood pressure ( SBP ) were measured, and the kidney hypertrophy index(KI) was calculated. The renal pathological sections of rats were observed. The expression of TGF-[31 and BMP-7 in renal tissue of rats in each group was detected by immunohistochemistry. Results There were significant differences in 24 h food intake, water intake, urine volume, UPro, UAlb, UOP, BUN, Scr, TG, UA, SBP, renal weight, KI, TGF-β1 and BMP-7 between model group and normal control group (P 〈 0. 05 ) , and the pathological changes of model group were obvious. After treatment, the 24 h food intake, water intake, urine volume, UPro, UAlb, UN, Scr, UA, TG,Ucr,SBP,kidney weight,KI and the expression of TGF-β1 in irbesartan group and Ernilia sonchifolia group were lower than those in model group( P 〈 0. 05 ) , while the expression of UOP and BMP-7 in irbesartan group and Ernilia sonchifolia group was higher( P 〈 0. 05 ).
