摘  要:目的观察追风透骨胶囊用于脊髓损伤模型大鼠的最优浓度及其对神经元的保护作用,探讨追风透骨胶囊降低损伤后神经元的炎症与凋亡水平的具体机制。方法将健康SD雌性大鼠随机分成3组,分别为脊髓损伤组、治疗组、假手术组。脊髓损伤组:应用经典的Allen's法制造损伤模型。治疗组:腹腔注射20 mg/kg追风透骨胶囊溶液。造模成功后,于术后第1、3、7、14、28天分别应用行为学试验(Basso、Beattie、Bresnahan locomotor rating scale,BBB评分)检测运动功能恢复情况。随后用尼式染色观察神经脊髓前角运动神经元数量。应用Western blot观察β-catenin、LRP-6、active Caspase-3表达。结果与脊髓损伤组比较,治疗组BBB评分在第7、14、28天明显升高(P <0.05,P <0.01);同时,与脊髓损伤组比较,治疗组β-catenin、LRP-6蛋白表达被激活(P <0.01),active Caspase-3表达水平明显降低(P <0.01)。结论脊髓损伤后,追风透骨胶囊可能通过β-catenin信号通路抑制损伤神经元产生的凋亡,从而对脊髓神经元产生保护作用。Objective To observe the best concentration of Zhuifengtougu capsule(ZFTG) in the treatment of spinal cord injury model rats and the protective effect of ZFTG on neurons,and explore the mechanism of ZFTG in relieving the inflammation and apoptosis of neurons. Methods The healthy female SD rats were randomly divided into three groups:spinal cord injury group, treatment group and sham group. Rats in spinal cord injury group were treated with the classic Allen's method to make the SCI model; rats in treatment group were injected intraperitoneally with 20 mg/kg ZFTG solution. The recovery of motor function was detected by the behavioral test (Basso, Beattie, Bresnahan locomotor rating scale and BBB scores) at 1,3,7,14 and 28 d after operation. The number of motor neurons in the anterior horn of the spinal cord was observed by Nissl staining. The expression of β-catenin,LRP-6 and active Caspase-3 was observed by Western blot. Results The BBB score of treatment group was higher than that of spinal cord injury group at the 7th, 14th and 28th day( P 〈 0. 05 or P 〈 0. 01 ). Compared with spinal cord injury group,the β-catenin and LRP-6 protein in treatment group was increased(P 〈 0. 01 ) , while the expression of active Caspase-3 was decreased( P 〈 0. 01 ). Conclusion ZFTG can inhibit the apoptosis of injured neurons via β-catenin signaling pathway after SCI, so as to protect spinal neurons.
