低氧诱导因子与高原老年骨性关节炎相关性的初步研究

摘　　要：[目的]检测低氧诱导因子1α(HIF-1α)、低氧诱导因子2α(HIF-2α)、促红细胞生成素(EPO),在高原骨性关节炎(OA)患者软骨和血清中的表达水平。[方法]选择高原地区骨关节炎患者11例(A组),亚高原地区骨性关节炎患者12例(B组),亚高原地区正常软骨9例(C组),分别采集血清及关节软骨行ELISA和免疫组化实验,监测HIF-1α、HIF-2α、EPO在血清和软骨中的表达情况。[结果] HIF-1α和HIF-2α在血清中的表达三组间差异无统计学意义,EPO A组高于B组及C组(P<0.05),B组与C组组间差异无统计学意义(P>0.05);HIF-1α在软骨中表达A组高于C组,而HIF-2α的表达A组和B组均高于C组(P<0.05),其中HIF-2αA组高于B组(P<0.05),EPO虽然在三组之间也呈现出A组高于B组及C组,但三组间差异无统计学意义。[结论] HIF-1α和HIF-2α与OA的发病相关,且有可能在高原OA患者发病中发挥重要作用。EPO虽然在世居高原人群血清中表达增加,但与OA发病无明显相关。[Objective] To detect the expression level of hypoxia inducible factor 1 alpha （HIF-1α）, hypoxia inducible factor 2 alpha （HIF-2α） and erythropoietin （EPO） in cartilage and serum from patients with osteoarthritis （OA）. [Methods] This study included 11 patients with OA in plateau region （group A）, 12 patients with OA in subtropical region （group B） and 9 patients with normal cartilage in subtropical region （gToup C）. The serum and articular cartilage were harvested to detect the expression level of HIF-1α, HIF-2α and EPO by ELISA and immunohistochemical assay. [Results] The expression of HIF-1α and HIF-2α in serum had no significant differences among the three groups （P〉0.05）. Regarding to EPO, the group A was statistical- ly higher than group B and C （P〈0.05）, despite the fact that no statistical difference was noted between group B and group C （P〈 0.05）. The expression of HIF-1α in cartilage tissue in the group A was higher than those in group C, and the expression of HIF- 2α in the group A and group B were significantly higher than those in group C （P〈0.05）. The expression of HIF-2α in cartilage tissue in the group A was higher than the group B （P〈0.05） . Although the EPO in the group A was slightly higher than those in the group B and group C, there was no a statistical difference among the three groups （P〉O.05） . [Conclusion] The HlF- l alpha and HIF-2 alpha is related to OA, which might play an important role in the pathogenesis of OA in plateau. Ahhough the erythro- poietin is of high expression in the serum of the plateau population, it might not be significantly related to the pathogenesis of OA.
