摘 要:建立妇女痛经颗粒中丹参酮ⅡA的含量测定方法。采用HPLC法,色谱柱:Diamonsil C18色谱柱(250mm×4. 6mm,5μm),以甲醇-水(75:25)为流动相,流速为1. 0mL·min^(-1),柱温35℃,UV检测波长为270nm,进样量为10μl,测定7批妇女痛经颗粒中丹参酮ⅡA的含量。在此色谱条件下,丹参酮ⅡA可与其他成分较好的分离,峰形对称,丹参酮ⅡA对照品在0. 6128~76. 6μg·mL^(-1)范围内与峰面积呈良好的线性关系(R^2=0. 9999)。样品平均回收率为97. 56%,RSD为1. 25%,精密度、稳定性、重复性的RSD均〈2%。该方法灵敏快速,操作简便,重现性好,可用于测定妇女痛经颗粒丸中丹参酮ⅡA的含量。To establish the assay of Tanshinone Ⅱ A in Funvtongjing Particles by high performance liquid chromatography ( HPLC ). Seven batches of Funvtongjing Particles were determined for the content of Tanshinone Ⅱ A by HPLC. The samples were separated on Diamonsil C18 (250mm × 4.6mm ,5μm) at:the column temperature was 35 ℃ , with the mobile phase composed of Methanol -water(75:25 ) at a flow of 1.0mL ·min-1. The detection wavelength was 270 nm and the sampling volume was 10 μl. It was found that Tanshinone Ⅱ A eould be isolated from the other components and the type of peak was symmetrical. Tanshinone Ⅱ A showed a good liner relationship with the peak area at the rage of 0.6128 - 76.6μg · mL-1 of Tanshinone Ⅱ A (R2 = 0. 9999). The average reeovelT of Tanshinone Ⅱ A was 97.56% and RSD was 1.25% , the RSDS of precision, stability, repeatability were less than 2%. This method was shown to be suitable for the determination of the content of Tanshinone Ⅱ A in Funvtongjing Particles.
