摘 要:目的:观察通秘合剂对大鼠结肠神经节细胞数目及Cajal间质细胞的影响,探讨通秘合剂治疗结肠慢传输型便秘(Slow transit constipation,STC)的作用机制。方法:将50只大鼠适应性喂养1周后,随机选取10只为空白组,其余大鼠按照复方地芬诺酯灌胃方法制备STC模型。将造模成功的36只大鼠随机分为模型组、对照组和治疗组,每组各12只。空白组及模型组给予生理盐水10 mL·(kg·d)^(-1)灌胃,治疗组给予通秘合剂10 mL·(kg·d)^(-1)灌胃,对照组给予莫沙必利混悬液10 mL·(kg·d)^(-1)灌胃。观察各组大鼠的一般情况、粪便含水量、肠推进进功能及神经节细胞和Cajal间质细胞的数量和结构。结果:造模组大鼠粪便水量较空白组明显下降,差异具有统计学意义(P <0.05)。给药后,治疗组和对照组粪便含水量高于模型组,差异具有统计学意义(P <0.05);与空白组比较,粪便含水量差异无统计学意义(P>0.05)。对照组、治疗组神经节细胞数目较模型组明显增多,治疗组神经节细胞数量上升明显多于对照组,差异具有统计学意义(P <0.05)。模型组ICC阳性表达明显低于空白组,对照组和治疗组多于模型组,治疗组有明显多于对照组,差异均有统计学意义(P <0.05)。结论:通秘合剂能够改善STC大鼠结肠神经节细胞及Cajal间质细胞的数量和结构。Objective: To observe the influence of Tong Mi Mixture( TMM) on the number of colon ganglion cells and Cajal mesenchymal cells in rats,and to explore the mechanism of TMM in the treatment of colon slow transit constipation( STC).Methods: After feeding adaptively 50 rats for one week,10 rats were randomly selected into the normal group.The remaining rats were prepared into STC models according to method of compound diphenoxylate for gavage.Then 36 rats which had been modeled were randomly divided into the model group,the control group and the treatment group,with 12 rats in each group.Results: The ones in the normal group and the model group were given 10 mL·(kg·d)-1 of normal saline for gavage.The ones in the treatment group were given 10 mL·(kg·d)-1 of TMM for gavage,and the ones in the control group were given 10 mL·(kg·d)-1 of the mosapride suspension for gavage.Then the general condition,fecal water content,intestinal propulsion function,the number and structure of ganglion cells and Cajal interstitial cells in each group were observed.Results: The fecal water volume of the rats in the model group was significantly lower than that in the blank group,and the difference was statistically significant( P 〈 0.05).After administration,the fecal water content of the treatment group and the control group was higher than that of the model group,the difference was statistically significant( P 〈 0.05).Compared with the blank group,there was no significant difference in the water content of the feces( P 〉 0.05).The number of ganglion cells in the control group and the treatment group increased significantly than that in the model group,and the number of ganglion cells in the treatment group increased significantly more than that in the control group,with a statistically significant difference( P 〈 0.05).The positive expression of ICC in the model group was significantly lower than that in the blank group,and that of the control group and the treatment group were m
