摘 要:目的:分析秋龙筋降糖中药制剂对链脲佐菌素(streptozocin,STZ)诱导糖尿病模型小鼠降低血糖的作用与机制。方法:取40只小鼠随机分为对照组、模型组、阳性组及观察组,模型组、阳性组及观察组采用STZ造模。造模成功后每组随机选6只小鼠,观察组给予秋龙筋降糖中药制剂干预,阳性组给予二甲双胍干预,对照组及模型组给予生理盐水。每周检测血糖,4周后检测血清中白细胞介素-2(interleukin-2,IL-2)、IL-4、IL-10及γ干扰素(interferon-γ,IFN-γ)水平,并分析肝脏组织中超氧化物歧化酶(superoxide dismutase,SOD)及脂质过氧化物(lipid peroxidase,LPO)水平。结果:造模后,模型小鼠空腹血糖水平均显著高于正常小鼠(P<0.05),且随着时间延长,小鼠空腹血糖也显著升高(P<0.05);治疗前模型组、阳性组、观察组小鼠血糖比较,差异无统计学意义(P>0.05),但显著高于对照组(P<0.05)。治疗后,阳性组及观察组小鼠血糖水平均明显降低,且差异有统计学意义(P<0.05);阳性组及观察组小鼠血清中IL-2、IFN-γ及IFN-γ/IL-4水平明显低于模型组,IL-10水平明显高于模型组,差异有统计学意义(P<0.05);阳性组明显优于观察组,差异有统计学意义(P<0.05);模型组小鼠LPO水平显著高于阳性组及观察组(P<0.05),SOD水平明显低于阳性组及观察组,差异有统计学意义(P<0.05),阳性组显著优于观察组(P<0.05)。结论:秋龙筋降糖中药制剂对STZ诱导糖尿病模型小鼠有显著治疗作用,可有效调控机体Th1细胞极化程度,抑制Th1细胞免疫功能,减轻胰岛β细胞的破坏作用,进而有效降低血糖水平。Objective:To analyze the effect and mechanism of Qiulongjin Jiangtang TCM Preparation on blood glucose lowering in- duced by streptozocin (STZ) in diabetic mice. Methods:Forty mice were randomly divided into control group,model group,posi- tive group and observation group. The model group, positive group and observation group were modeled by STZ. After successtul modeling,6 mice were randomly selected from each group. The observation group was given the intervention of Qiulongjin Jiangtang TCM Preparation;the positive group was given metformin intervention;the control group and the model group were given normnal sa- line. Blood glucose was measured weekly, and serum levels of interleukin-2 ( IL-2 ), IL-4, IL- 10, interferon-γ ( IFN-γ), superoxide dismutase (SOD) and lipid peruxidase (LPO) levels in liver tissue were measured and analyzed after 4 weeks. Results :After mod- el establishment, the fasting blood glucose level of the model mice was significantly higher than that of normal mice ( P 〈 0. 05 ), and the fasting blood glucose of the mice was also significantly increased with time ( P 〈 0.05 ). There was no siznificant difference in blood glucose between the model group, the positive group and the observation group (P 〉 0.05 ), but that was significantly higher than that of the control group (P 〈 0.05 ). After treatment, the blood glucose levels of the positive and observation groups were significantly lower, and the difference was statistically significant ( P 〈 0.05 ). The level of the serum IL-2, IFN-γand IFN-γ IL-4 in the positive and observation groups was significantly lower than that of the model group, and the IL-10 level was significant- ly higher than that of the model group.
