丹溪痛风胶囊对类风湿性关节炎大鼠血清IL-17和IL-23水平的影响

摘　　要：目的:观察丹溪痛风胶囊对类风湿性关节炎(rheumatoid arthritis,RA)大鼠的治疗作用及对血清白细胞介素-17(interleukin-17,IL-17)和IL-23水平的影响。方法:32只大鼠随机分成正常组,模型组,丹溪痛风胶囊低剂量组、高剂量组。除正常组外,其余各组采用牛Ⅱ型胶原诱导法制备类风湿性关节炎大鼠模型,各组分别灌胃给予相应药物。给药28 d后,检测各组大鼠足跖肿胀度和关节炎指数,显微镜下观察大鼠关节滑膜的病理变化,ELISA法检测血清中IL-12和IL-23的水平。结果:模型组大鼠足跖肿胀度和关节炎指数明显增高,与正常组比较,差异有统计学意义(P<0.01);模型组大鼠关节滑膜水肿、增生明显,并有大量炎细胞浸润;模型组大鼠血清中IL-12和IL-23的水平均显著升高,与正常组比较,差异有统计学意义(P<0.01)。丹溪痛风胶囊低剂量组和高剂量组大鼠的足跖肿胀度和关节炎指数均不同程度降低,与模型组比较,差异有统计学意义(P<0.01);丹溪痛风胶囊低剂量和高剂量均能明显改善模型大鼠关节滑膜的病理变化;丹溪痛风胶囊低剂量组和高剂量组大鼠血清中IL-12和IL-23的水平均明显降低,与模型组比较,差异有统计学意义(P<0.01)。结论:丹溪痛风胶囊通过下调血清中IL-12和IL-23的水平起到治疗RA的作用。Objective ：To observe the effects of Chinese medicine Danxi＇s gout capsule on expressions of IL-17 and IL-23 in colla- gen-induced arthritis （CIA） rats. Methods：Forty male SD rats were randomly divided into 4 groups：normal group, model control group, Danxi Gout Capsule low-dose treatment group and high-dose treatment group. After CIA was established in rats successfully, Danxi＇s gout capsule low-dose and high-dose treatment group were treated with Danxi＇s gout capsule by gavage,while the normal group and model control group were fed with the same amount of saline. Four weeks after the treatment, the rats were collected ser- um and sacrificed. The pathological of CIA joint synovial was observed by electron microscope. The expressions of IL-17 and IL-23 in serum were detected by ELISA. Results ： The degree of swelling of the toes and arthritis index of the model group were signifi- cantly increased. Compared with the normal group, the difference was statistically significant （P 〈 0.01 ）. The synovial edema and hyperplasia of the joints in the model group were obvious, and there were a large number of inflammatory cells and much infiltra- tion,and the levels of IL-12 and IL-23 in the serum of the model group were significantly increased （P 〈0. O1 ）. The toe swelling degree and arthritis index of Danxi Gout Capsule low-dose group and high-dose group were decreased to different degrees, corn-pared with that of the model group ,the difference was statistically significant （P 〈 0.01 ）. Danxi Gout Capsule low dose and high dose can significantly improve the pathological changes of joint synovial membrane in rats and the levels of IL-12 and IL-23 in the serum of Danxitongfeng capsule low dose group and high dose group were significantly lower than those of the model group （P 〈 0.01 ）. Conclusion：Danxi Gout Capsule can treat RA with significant clinical efficacy by down-regulating the levels of IL-12 and IL-23 in serum.
