双黄二仙汤对db/db小鼠肾脏的保护作用及对肾组织上皮细胞钙黏蛋白和α-肌动蛋白表达的影响

摘  要:目的:观察双黄二仙汤对db/db小鼠肾脏的保护作用及对肾组织上皮细胞上皮细胞钙黏蛋白和α-肌动蛋白表达的影响。方法:选取12周龄雄性C57BLKs/Jdb/m小鼠10只,以及使用链脲佐菌素35~40 mg·kg-1快速注射造模的12周龄雄性C57BLKs/Jdb/db小鼠20只,30只小鼠饮水不限,均给予饲喂颗粒饲养,然后经过1周观察进入实验。将20只db/db小鼠随机平均分为双黄二仙汤组和模型组。双黄二仙汤组每日予以双黄二仙4.5 g(生药)·kg-1灌胃;模型组予以等量生理盐水灌胃处理;正常对照组予以相同体积的生理盐水灌胃处理。观察不同处理方式对db/db小鼠肾脏功能的保护作用,以及对上皮细胞钙黏蛋白与α-肌动蛋白表达的影响。结果:实验开始时正常对照组小鼠的体质量明显低于模型组与双黄二仙汤组(P<0.05),模型组与双黄二仙汤组小鼠体质量无明显差异(P>0.05);灌胃处理后到结束期间,3组小鼠的体质量均上升,而模型组小鼠体质量上升程度明显高于正常对照组与双黄二仙汤组(P<0.05),双黄二仙汤组小鼠体质量上升程度高于正常对照组(P<0.05)。灌胃后模型组小鼠24小时尿蛋白定量、尿素氮(urea nitrogen,BUN)、肌酐(creatinine,Cr)、尿酸(Uric Acid,UA)、空腹血糖(fasting blood-glucose,FBG)、三酰甘油(triacylglycerol,TG)、总胆固醇(total cholesterol,TC)、糖基化终末产物(advanced glycation end products,AGEs)表达水平明显高于双黄二仙汤组与正常对照组(P<0.05),而双黄二仙汤组小鼠各指标的表达水平高于正常对照组(P<0.05)。灌胃后模型组小鼠上皮细胞钙黏蛋白阳性表达情况明显低于正常对照组与双黄二仙汤组,α-肌动蛋白表达水平高于正常对照组与双黄二仙汤组(P<0.05),双黄二仙汤组小鼠上皮细胞钙黏蛋白阳性表达情况低于正常对照组,而α-肌动蛋白高于正常对照组(P<0.05)。结论:双黄二仙汤对db/db小鼠肾脏功能具有一定的保护作用,�Objective:To investigate the protective effect of Shuanghuang Erxian Decoction on kidney of db/db mice and its effect on the expression of E-cadherin andct-aetin in renal tissue. Methods:30 rats, namely, ten male C57BLKs/Jdb/m mice of 12 weeks old and 20 male C57BLKs/Jdb/db mice of 12 weeks old, were injected rapidly with streptozotocin 35 -40 mg · kg^-1. The mice were given water and were fed with pellets, and then were observed for 1 week. Twenty db/db mice were randomly divided into Shuanghuang Erxian Decoction group and model group. Shuanghuang Erxian Decoction group was given Shuanghuang Erxian 4.5 g (raw medicine) · kg^-l per day;the model group was treated with the same amount of normal saline. The normal control group was given the same volume of normal saline. The protective effects of different treatments on renal function of db/db mice and the effects of cadherin and oL-actin expression on epithelial cells were observed. Results : The body weight of the normal control group was significantly lower than that of the model group and Shuanghuang Erxian Decoction group (P 〈 0.05 ). There was no signifi- cant difference in the body weight between the model group and Shuanghuang Erxian Decoction group (P 〉 0.05 ). After the stom- ach treatment to the end, the body weight of the three groups of mice increased, while the body weight of the model group was sig- nificantly higher than that of the normal control group and Shuanghuang Erxian Decoction group ( P 〈 0.05 ). The degree of quality increase was higher than that of the normal control group ( P 〈 0.05 ). The 24 hours urine protein quantitation, urea nitrogen (BUN), creatinine (Cr), uric acid (UA) ,fasting blood-glucose (FBG), triacyl in mice in the model group after intragastric ad- ministration, the expression levels of triacylglycerol (TG), total cholesterol (TC) and advanced glycation end products (AGEs) were significantly higher than those of Shuanghuang Erxian Decoction group and normal c
