摘 要:为了解我国Ⅰ群禽腺病毒(FAdV-Ⅰ)主要流行血清型hexon蛋白序列和抗原表位的差异,对43株FAdV-Ⅰ分离株的hexon进行了序列和遗传进化分析,利用DNAStar中Protean预测了4个流行血清型代表株hexon蛋白的抗原表位。结果显示:同型分离株间hexon氨基酸序列同源性高,其中FAdV-4分离株的氨基酸序列同源性为100%。hexon蛋白氨基酸序列在第132~289位、第363~507位和第793~878位存在3个主要可变区,分别对应于hexon的L1、L2和L4区。在FAdV-11中,分离株QDPD100808和YTLY081010氨基酸序列在第97-138位区段发生缺失,分离株HNQX120913在第693-720位突变为HVQEHERALRHVHQLARERPVASTQPVS。预测到FAdV-4、FAdV-8a、FAdV-8b和FAdV-11的代表株hexon蛋白分别有40、37、38和37个抗原表位,FAdV-4的特异性抗原表位最多,为16个,FAdV-8a、FAdV-8b和FAdV-11特异性抗原表位较少,分别为3个、6个和3个。研究结果为我国FAdV-Ⅰ主要流行血清型基于hexon基因的诊断提供了理论依据。To understand the differences of hexon protein sequence and antigen epitope of the main epidemic serotype of fowl adenovirus group Ⅰ(FAdV-Ⅰ)in China,the hexon sequence and phylogenetic of 43 isolates of FAdV-Ⅰ were analyzed, and the antigen epitopes in hexon of 4 main epidemic serotypes were predicted by Protean of DNAStar. The results showed that the amino acid homology of hexon was high among the same serotype isolates, with 100% amino acid homology of FAdV-4. There were three major variable regions (140 to 289, 363 to 507 and 793 to 878)of hexon protein, which correspond to the L1, L2 and L4 regions of hexon, respectively. Among FAdV-Ⅱ isolates there was a deletion region (97 to 138)both in QDPD100808 and YTLY081010, moreover there was a mutation region (693 to 720) in HNQX120913, which sequence was HVQEHERALRHVHQLARERPVASTQPVS. It was predicted that the hexon protein of FAdV-4, FAdV-8a, FAdV-Sb and FAdV-11 had had 16 specific antigen epitopes, while FAdV-8a, FAdV-8b respectively. This study provided a theoretical basis for the China based on the hexon gene. 40, 37, 38 and 37 antigen epitopes, respectively. FAdV-4 and FAdV-11 had 3, 6 and 3 specific antigen epitopes, diagnosis of the main epidemic serotype of FAdV-Ⅰ in China based on the hexon gene.
