摘  要:R-藻红蛋白(R-PE)是一种新型荧光标记探针,具有优良的荧光特征。本实验采用异双功能交联试剂SPDP和SMCC分别活化R-PE和CD4/CD8单克隆抗体,再将活化后的R-PE和CD4/CD8单克隆抗体以一定比例混合,使之发生交联反应,交联产物经SephacrylS-300柱分离纯化。结果显示:用R-PE标记的抗CD4/CD8单抗,经流式细胞仪(简称FACS)分析可以检测正常人外周血淋巴细胞表面CD4抗原和CD8抗原的表达,且R-PE标记的抗CD4/CD8单抗特异性保持完好,荧光强度较高,可以形成单色或和用其他荧光染料标记的CD系列单抗组成双色、多色的荧光试剂,应用于流式细胞仪检测分析。R-phycoerythrin (R-PE) is a novel fluorescent probe, with excellent fluorescence characteristics. In this study, the R-PE and the anti-CD4/CD8 MCAb were activated by heterobifunctional reagent SPDP and SMCC respectively. And then the activated R-PE and anti-CD4/CD8 MCAb were conjugated by being mixed in a ratio of 1:1. The conjugate was purified by Sephaeryl S-300HR gel filtration, The results showed that the R-PE conjugate is a useful fluorescent reagent for analysis CD4/CD8 on human peripheral blood lymphocytes. And the fluorescein-labeled McAbs maintained good specificity and high intensity. It is applied to flow cytometry analysis.
