外周血DNT细胞在支气管哮喘急性加重的临床意义

摘　　要：目的:研究支气管哮喘急性加重患者外周血T淋巴细胞亚群中双阴性T淋巴细胞(DNT)的变化及其临床意义。方法:收集我院2015年4月至2017年11月收治的101例支气管哮喘急性加重患者作为哮喘组,再将哮喘组根据哮喘发作的程度分为轻、中、重、危重4组,选择同期健康体检者90例作为对照组。采用FC500流式细胞仪检测受检者的外周血DNT细胞的表达水平,并对数据进行统计学分析。结果:对照组DNT细胞数量表达显著高于支气管哮喘急性加重组,差异有统计学意义(P <0. 05),其中重度组DNT细胞数表达亦明显低于轻度组及中度组,差异具有统计学意义(P <0. 05)。结论:支气管哮喘急性加重患者外周血DNT细胞低表达,且DNT细胞低表达的程度与支气管哮喘急性发作的严重程度相关。提示外周血DNT细胞低表达在支气管哮喘急性加重期中起到一定病理生理作用,并可以通过测定DNT细胞表达水平判定支气管哮喘急性加重的严重程度。Objective： To study the change and clinical significance of double-negative T-cells（ DNT） in peripheral T lymphocyte subsets of patients with acute exacerbation of bronchial asthma. Methods： A total of 101 cases of patients with acute exacerbation of bronchial asthma admitted to our hospital from April 2015 to November2017 were selected as asthma group,which was subdivided into mild group,moderate group,severe group and critical group according to the degree of asthmatic attack,and 90 healthy cases in the corresponding period were selected as control group. FC500 flow cytometry（ FCM） was adopted to detect the expression level of DNT cells in the peripheral blood of the objects,and on the data were statistically analyzed. Results： DNT cell quantity of patients in the control group was significantly higher than that of acute exacerbation of bronchial asthma（ P〈0. 05）,and the difference was statistically significant; DNT cell quantity of patients in severe group was significantly lower than that in mild group and moderate group（ P〈0. 05）,and the difference was statistically significant. Conclusion： The expression of DNT cells in peripheral blood of patients with acute exacerbation of bronchial asthma is low,and the degree of low expression of DNT cells is correlated with the severity of acute attack of bronchial asthma,suggesting that the low expression of DNT cells in peripheral blood playssomepathophysiological effect in acute exacerbation of bronchial asthma,and the severity of acute exacerbation of bronchial asthma can be determined by measuring the expression level of DNT cells.
