摘　　要：目的:研究针刺预处理对缺血再灌注损伤大鼠心肌内质网应激的影响。方法:48只Wistar雄性大鼠随机分为4组:假手术组(Sham)、缺血再灌注组(I/R)、电针干预组(EP)、电针对照组(EC)。建立心肌缺血再灌注模型,EP组于手术前1周即每日给予电针"内关穴"(P6)处理,而EC组则予电针"外关穴"(SJ5)处理,免疫印迹法与免疫组化法检测内质网应激相关蛋白及p-Akt的表达。结果:I/R组心肌细胞内质网相关蛋白Caspase-12、Chop较Sham组升高,EP组内质网相关蛋白Caspase-12、Chop较I/R组下降;EP组显著上调心肌组织p-Akt的表达,与I/R组相比具有显著统计学意义(P<0.05);同时与I/R组相比,电针"外关穴"并未发现p-Akt的激活以及Caspase-12、Chop表达的下调。结论:针刺预处理通过抑制心肌缺血再灌注后的内质网应激,其机制可能与激活Akt磷酸化有关。Objective： To research the effect of acupuncture preconditioning on endoplasmic reticulum stress in rats with myocardial ischemia-reperfusion injury. Methods： 48 Wistar male rats were randomly divided into4 groups：sham operation group（ Sham）,ischemia reperfusion group（ I/R）,electro-acupuncture group（ EP）and electroacupuncture control group（ EC）. The model of myocardial ischemia reperfusion was established.Group EP was treated with electro-acupuncture ＂Neikuan point＂（ P6） daily 1 week before operation,while group EC was treated with electro-acupuncture of＂Waikuan point＂（ SJ5）,and the expressions of endoplasmic reticulum stress related protein and p-Akt were detected by immunoblotting and immunohistochemistry. Results： The endoplasmic reticulum related proteins Caspase-12 and Chop in the I/R group were higher than those in the Sham group. The endoplasmic reticulum related proteins,Caspase-12 and Chop in the EP group were lower than those in the I/R group,and the EP group significantly up-regulated the expression of p-Akt in rats＇ myocardium,it was significant statistical significance while compared with the I/R group（ P〈0. 05）.Compared with group I/R,the activation of p-Akt and the down-regulation of Caspase-12 and Chop expression were not found in EA group. Conclusion： Acupuncture preconditioning inhibited endoplasmic reticulum stress after myocardial ischemia-reperfusion,and its mechanism may be related to the activation of p-Akt.
