桂枝茯苓丸对盆腔炎模型大鼠子宫、卵巢组织MMP-9表达影响的实验研究

摘　　要：目的:观察桂枝茯苓丸对盆腔炎模型大鼠子宫、卵巢组织中基质金属蛋白酶-9(MMP-9)表达的影响,旨在为临床运用活血化瘀药物桂枝茯苓丸治疗盆腔炎性疾病提供参考。方法:144只大鼠随机分为空白组、假手术组、模型组、甲硝唑组和桂枝茯苓丸组;运用"混合细菌接种"法进行造模,造模成功后第7 d给予相关药物治疗21 d。记录大鼠一般情况,免疫组化法测定各组大鼠子宫、卵巢组织MMP-9的表达。结果:模型组大鼠一般情况、体质量与正常对照组比较差异有统计学意义(P<0.05);桂枝茯苓丸组与甲硝唑组比较有显著统计学意义(P<0.01)。模型组大鼠子宫、卵巢组织MMP-9表达较正常组明显升高,阳性对照组和桂枝茯苓丸组MMP-9表达较模型组显著降低(P<0.01),桂枝茯苓丸组表达低于阳性组(P<0.05)。结论:桂枝茯苓丸能够降低盆腔炎大鼠子宫和卵巢组织的MMP-9表达,减轻盆腔组织的粘连。Objective： To observe the effect of Guizhi Fuling Pill on metalloproteinase（ MMP-9） expression in uterine and ovarian tissue of pelvic inflammation rats,in order to provide reference for clinical application of Guizhi Fuling Pill in the treatment of pelvic inflammation disease. Methods：140 rats were randomly divided into normal,sham,model,positive and Guizhi Fuling Pill groups,models with pelvic inflammation were made by mixed bacterial inoculation method,each group was given the corresponding distilled water or medicine from the 7^（th） day to the 21^（st） day. The general condition of rats were recorded. The expression of MMP-9 in uterus and ovary tissue of each group was determined by immunohistochemistry. Results： The general condition and body weight of the model group rats were significantly different from those of the normal control group（ P〈0. 05）; the Guizhi Fuling Pill group had a statistically significant difference with the metronidazole group（ P〈0. 01）. The expression of MMP-9 in the uterus and ovarian tissues of the model group was significantly higher than that of the normal group. The expression of MMP-9 in the positive control group and the Guizhi Fuling Pill group was significantly lower than that in the model group（ P〈0. 01）,and the expression in the Guizhi Fuling Pill group was lower than that of the positive group（ P〈0. 05）. Conclusion：Guizhi Fuling Pills can decrease MMP-9 expression in uterine and ovarian tissue of pelvic inflammation rats to reduce the pelvic tissue adhesion.
