虎刺提取物对佐剂性关节炎大鼠治疗作用及机制探讨

摘　　要：目的:通过佐剂性关节炎大鼠(AA大鼠)模型探究虎刺水提取和醇提物对类风湿性关节炎(RA)的治疗作用及其机制。方法:采用弗氏完全佐剂诱导大鼠关节炎,大鼠随机分成9组:正常对照组、模型对照组、阳性药对照组(甲氨蝶呤3 mg/kg)、虎刺水提物低剂量(0.4 g/kg)、中剂量(0.8 g/kg)、高剂量(1.6 g/kg)组和虎刺醇提物低剂量(0.4 g/kg)、中剂量(0.8 g/kg)、高剂量(1.6 g/kg)组,每组10只。观察大鼠体质量、足肿胀度、关节炎评分指数、免疫器官(胸腺、脾脏)指数及血清中细胞因子(TNF-α、IL-1β、IL-6和IL-10)的变化。结果:在给药第14 d后,虎刺水提物低剂量、中剂量、高剂量组和醇提物中剂量、高剂量组能抑制AA大鼠足肿胀;虎刺水提物低剂量、中剂量、高剂量组和醇提物高剂量组关节炎指数低于模型对照组;虎刺水提物和醇提物各组别的胸腺指数均高于模型对照组;虎刺水提物中剂量、高剂量)组和醇提物高剂量组均能改善降低炎性因子TNF-α、IL-1β、IL-6水平,升高IL-10水平。结论:虎刺水提物和醇提物能有效改善AA大鼠关节炎症,提示虎刺具有一定抗类风湿性关节炎(RA)作用。该抗RA作用可能与调节细胞因子水平和免疫保护作用相关。Objective： To observe the therapeutic effect of Damnacanthus indicus extracts on rheumatoid arthritis（ RA） in adjuvant arthritis（ AA） rats and explore the mechanism. Methods： Arthritis models were induced by Freund ＇s complete adjuvant. Nine groups including normal control,model control,positive control（dexamethasone,5 mg/kg）,Damnacanthus indicus aqueous extract（0. 4,0. 8,1. 6 g/kg） and alcohol extract（0. 4,0. 8,1. 6 g/kg） groups were set up. The weight,paw volume,arthritis index score,thymus and spleen indexes,serum levels of TNF-α,IL-1β,IL-6 and IL-10 were detected. Results： The rats＇ paw volume of Damnacanthus indicus aqueous extract group（0. 4,0. 8,1. 6 g/kg） and alcohol extract group（ 0. 8,1. 6 g/kg） were reduced. The rats＇ arthritis index score of Damnacanthus indicus aqueous extract group（ 0. 4,0. 8,1. 6 g/kg） and alcohol extract group（1. 6 g/kg） was much lower than that of model. The thymus index of Damnacanthus indicus aqueous extract group and alcohol extract group was higher than that of model. The serum levels of TNF-α,IL-1β,IL-6 of Damnacanthus indicus aqueous extract group（ 0. 8,1. 6 g/kg） and alcohol extract group（1. 6 g/kg） were lowered and IL-10 level was raised. Conclusion： Damnacanthus indicus aqueous extract and alcohol extract can improve inflammatory condition of AA rats,which suggested that Damnacanthus indicus showed anti-rheumatoid arthritis（ RA） effect. The anti-rheumatoid arthritis（ RA） effect was possibly related to cytokine levels and immunoprotection.
