摘 要:目的:观察青少期应激结合成年后诱导抑郁大鼠行为学、海马CA1区5-HT_(1A)R表达的变化及加味四逆散干预作用。方法:Wistar大鼠随机分成空白组、模型组、加味四逆散组、氟西汀组4组,每组33只;组内随机再分为青少期、成年期、成年后3小组,每小组11只。选取44、56、78 d3个时间点作为青少期、成年期、成年后3个阶段的取材时间点。采取慢性不可预知应激模型,第21-44天、第57-78天造模及给药; 44~55天正常喂养不予造模及给药。观察大鼠的一般情况与旷场实验情况,采用免疫组织化学染色法观察海马CA1区5-HT1A R阳性细胞的分布变化。结果:各时期模型组大鼠一般状况较差,加味四逆散组及氟西汀组有明显改善。旷场实验活跃度上,青少期与成年后模型组显著降低(P 〈0. 05),加味四逆散组与氟西汀组活跃程度显著升高(P 〈0. 01、P 〈0. 05)。总区路程变化结果中,成年期模型组大鼠有显著降低(P 〈0. 05),成年期及成年后加味四逆散组有显著提升(P 〈0. 05、P 〈0. 01);且在成年后与氟西汀组对比有显著差异(P 〈0. 01)。各组大鼠海马CA1区5-HT_(1A)R平均光密度值组内比较,青少期与成年后组模型组有显著降低(P 〈0. 05,P 〈0. 01);组间比较,成年后模型组、氟西汀组呈显著上升(P 〈0. 01,P 〈0. 05);加味四逆散组有上升趋势。结论:在青少期受到慢性不可预知应激的大鼠可产生抑郁样行为,成年后继续施加刺激则会进一步加重抑郁状态,海马内5-HT1AR改变可能是加味四逆散改善应激大鼠抑郁样行为的重要通路。Objective:To preliminarily observe the behavior and the content of 5-HT1AR in the hippocampus CA1 of adolescent stress combining adult induced depression rats and the intervention effect of Jiawei Sini Powder. Methods:Wistar rats were randomly divided into blank group,control group,Jiawei Sini Powder( JWSNP) group,fluoxetine group,33 rats in each group and the groups were randomly subdivided into youth stage,adult stage,after adulthood stage,11 rats in each stage. 44,56 and 78 days were selected as the sampling time points of youth stage,adult stage,after adulthood stage. CUMS model was used. From 21 d to 44 d and from 57 d to 78 d,we made the model and gave the medicine. But from 44 d to 55 d,we gave the normal feed and drug delivery. Then we observed the the behavior,open field test,the distribution of 5-HT1AR positive cells in hippocampus CA1 by immunohistochemical staining and the changes of5-HT_(1A)RmRNA expression in rats by RT-PCR technique. Results:The general condition of the rats in the control group was poorer,but the JWSNP group and fluoxetine group's improved obviously. In the activity of open field test,control group's decreased significantly in youth stage and after adulthood stage( P〈0. 05) and JWSNP group and fluoxetine group's improved obviously( P〈0. 01,P〈0. 05). In the Overall range variation,the adult rats in the control group had decreased( P〈0. 05). JWSNP group's was significantly improved in the adult stage and after adulthood stage( P〈0. 05 and P〈0. 01) and JWSNP group had significant differences compared with that of the fluoxetine group( P〈0. 01). Theaverage optical density of 5-HT1AR in hippocampus CA1 area of each group was compared and the control group's decreased significantly in youth stage and after adulthood stage( P〈0. 05,P〈0. 01).
