消痰散结方对原位移植胃癌裸鼠生长及血清IL-8、TGF-β的影响

摘　　要：目的：研究消痰散结方对原位移植胃癌裸鼠生长及血清IL-8、TGF-β的影响,探讨该方抑制胃癌转移机制。方法：建立裸鼠原位移植SGC7901胃癌模型,随机分为模型组、生理盐水组、消痰散结方组、5-氟脲嘧啶组。另取10只正常组做空白对照。观察裸鼠生长情况,计算抑瘤率;应用酶联免疫吸附测定法（ELISA）检测各组小鼠血清（IL-8、TGF-β）的浓度。结果：消痰散结方组和化疗组抑瘤率分别为41. 07%和60. 71%,与模型组及生理盐水组比较,差异有统计学意义（P 〈0. 05）。模型组较其他组血清IL-8、TGF-β浓度显著上升,中药组与模型组相比,显著下调血清IL-8、TGF-β的浓度,差异均有统计学意义（P 〈0. 05）,与化疗组相当,差异无统计学意义（P〉 0. 05）。结论：消痰散结方有较好抑瘤效果,能够下调原位移植胃癌裸鼠血清IL-8、TGF-β浓度,这可能是该方抑制胃癌转移的机制之一。Objective： To observe the inhibitory effect of Xiaotan Sanjie Formula（ XTSJF） and the influence on serum IL-8 and TGF-β in gastric cancer of nude mice and explore the possible mechanism of XTSJF in inhibiting the metastasis of the stomach cancer. Methods： SGC7901 gastric carcinoma orthotopic-transplanted model in nude mice was established. Forty nude mice were randomly divided into control group,normal sodium group,Xiaotan Sanjie Formula group and 5-fluorouracil（5-FU） group. In addition,ten nude mice were set as a blank-control group. The IL-8 and TGF-β serum concentrations of nude mice were measured with enzyme-linked immunosorbent assay（ ELISA）. Results： The inhibitory rates of XTSJF group and 5-FU group were 41. 07% and 60. 71% respectively. Compared with the control group and normal sodium group,the difference was statistically significant（ P〈0. 05）. XTSJF group significantly decreased the IL-8 and TGF-β serum concentrations compared with the control group（ P〈0. 05）,and there was no difference when compared with 5-FU group. Conclusion： XTSJF possesses anti-tumor effects on experimental tumor models and could down-regulate the IL-8 and TGF-β serum concentrations,which may be one of the mechanism of XTSJF anti-stomach carcinoma activity.
