摘  要:目的:观察藏药三果汤散对高原红细胞增多症模型大鼠抗氧化等指标表达的影响,探究三果汤散干预高原红细胞增多症模型大鼠的氧化应激作用机制。方法:雄性SD大鼠48只,每组8只,随机分为平原组,高原模型组,阳性对照组,三果汤高、中、低剂量组。将模型组和给药组置于5000米高原环境模拟舱内,40 d后测定大鼠血常规、血流变、SOD、MDA、EPO、ROS、HIF-1α等指标,探究三果汤散干预高原红细胞增多症大鼠氧化应激的作用机制。结果:与高原模型组相比,三果汤高剂量组和中剂量组的RBC、HGB和HCT显著降低,血液黏稠度也有显著改善,三果汤散高剂量组大鼠脑组织HIF-1α蛋白含量降低。与平原对照组相比,高原模型组大鼠血清SOD活性显著降低,MDA含量显著升高,脑组织HIF-1α蛋白含量显著升高,EPO浓度显著升高,EPO mRNA表达明显升高(P 〈0. 05)。结论:三果汤散具有明显的抗缺氧作用,能降低耗氧量、清除氧自由基、降低脂质过氧化反应、抑制缺氧造成的血流变改变、改善机体的能量代谢,这可能是其治疗高原红细胞增多症模型大鼠氧化应激损伤的作用机制。Objective: To observe anti-hypoxia pharmacological effects of tibetan medicine Sanguo Decoction/Powder of high altitude polycythemia rats model and explore its machanism. Method: Forty-eight SD rats were randomized into plain control group,plateau control group,positive medicine group,Sanguo Decoction high,middle and low doses groups. Put plateau control group and medication groups into the hypoxia environment simulated model. After 40 days,the blood routine,blood rheology,SOD,MDA,EPO,ROS and HIF-1α were measured,and we explored oxidative stress mechanism of Sanguo Decoction/Powder intervention on high altitude polycythemia rats model. Results:The blood indexes showed that the RBC,HGB,HCT of Sanguo Decoction groups significantly decreased compared with those of the plateau control group and blood viscosity was also significantly improved. The content of HIF-1α protein in brain tissue of rats was decreased in Sanguo Decoction high dose group. Compared with plain control group,plateau model could decrease the activities of SOD in mice,increase the contents of MDA,HIF-1α,EPO and EPOmRNA( all P〈0. 05).Conclusion: Sanguo Decoction/Powder has significant anti-anoxia effect. The oxidative stress mechanism of Sanguo Decoction/Powder may be related to decreasing consumption of oxygen,eliminating oxygen free radicals,reducing lipid peroxidation,inhibiting the changes of hemorheological and improving energy metabolism.
