摘 要:目的:通过网络药理学方法探究赤芍治疗糖尿病肾病的潜在作用机制。方法:通过中药系统药理学数据库及分析平台(TCMSP)筛选出赤芍的活性成分及靶点,挖掘TTD、Drug Bank、OMIM、GAD、Pharm GKB等数据库与糖尿病肾病的相关靶点,并用Cytoscape 3. 2. 1软件分别构建药物-靶点、疾病靶点网络图,利用网络拓扑分析出赤芍治疗糖尿病肾病的关键靶点,采用Clue GO信号通路富集分析赤芍治疗糖尿病肾病的潜在作用通路。结果:通过OB,DL条件筛选,得出27个入血活性成分,并预测出85个潜在靶点。通过筛选上述疾病基因相关数据库分析得出99个与糖尿病肾病发病相关的靶点。利用网络拓扑分析最终筛选出79个关键靶点,Clue GO富集分析得出赤芍治疗糖尿病肾病的关键靶点主要被富集在94个信号通路,涉及增殖、凋亡、炎症、代谢等生物学过程。结论:本研究结果初步验证和预测了赤芍对治疗糖尿病肾病的作用机制,并为进一步深入揭示其作用机制提供参考。Objective:To investigate the potential molecular mechanism of red peonyin the treatment of diabetic nephropathy based on network pharmacology. Methods:We screened the active compounds corresponding to red peony by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform. Then,we retrieved gene targets associated with diabetic nephropathy by the databases of TTD,Drug Bank,OMIM,GAD and Pharm GKB. Next,the drug-target interaction network as well as target-disease interaction network were constructed. And network topological analysis was used to determine the specific target of red peony in the prevention and treatment of diabetic nephropathy. Then,we performed enrichment analysis of the candidate targets of red peony using Clue GO. Results: There were 75 corresponding compounds retrieved in the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform. According to the values of OB and DL,a total of 27 candidate components were screened,which obtained 85 targets by using the related target prediction technique. There were 99 targets closely related to the development of diabetic nephropathy by screening in the disease-related database. Using the Cytoscape to construct and synthesize the protein-protein interaction network of active ingredients and diseases,79 key gene targets were screened out by network topological analysis. Using Clue GO analysis,the targets were mainly enriched in 94 signaling pathways,involving proliferation,apoptosis,inflammation,metabolism. Conclusion: The results preliminarily verified and predicted the mechanism of red peony on the treatment of diabetic nephropathy,and laid a good foundation for further study on the mechanism of action.
