摘 要:为了揭示绒毛白蜡种质之间的遗传多样性和亲缘关系,本试验选取50份绒毛白蜡种质,采用荧光标记AFLP技术,利用筛选出的9对Eco RⅠ/MseⅠ引物对绒毛白蜡种质进行遗传变异及遗传关系分析。结果表明,50个绒毛白蜡种质共扩增出18 451条条带,其中多态性条带17 301条,多态位点百分率高达93.75%;Nei's基因多样性指数平均为1.29,Shannon's多态性信息指数平均为0.30;各种质间的相似系数范围为0.458~0.741,当相似系数为0.62时,UPGMA分析将50个无性系分成9组,显示出绒毛白蜡种质资源遗传背景复杂,遗传多样性丰富。本研究为绒毛白蜡种质鉴定及资源保护提供分子证据,也为杂交育种的亲本选择及品种改良提供科学依据。In order to reveal the genetic diversity and the relationship between Fraxinus velutina Ton'., the genetic variation and genetic relationships of 50 Fraxinus velutina germplasms were analyzed by the fluorescent-AFLP technique with 9 EcoR I/Mse I primer combinations selected. This result showed that 50 Fraxinus velutina cultivars revealed a total number of 18 451 unambiguous bands, of which 17 301 were polymorphic and the percentage of polymorphism loci was 93.75%. On average, the Nei's gene diversity and Shannon's information index were 1.29 and 0.30, respectively. The similarity coefficient of 50 Fraxinus velutina cultivars ranged from 0.458 to 0.741. When the similarity coefficient was 0.62, these 50 Fraxinus velutina cultivars were divided into nine groups by UPGMA. The results showed that there were rich genetic diversity and complex genetic background in Fraxinus velutina germplasms resources. This study might provide molecular evidence for the germplasm identification and resource protection of Fraxinus velutina and also might provide a scientific basis for the parental selection and variety improvement of hybrid breeding.
