裸花紫珠颗粒体外抗肠道病毒71型及幽门螺杆菌的活性研究

摘　　要：目的：研究裸花紫珠颗粒体外抗肠道病毒71型（EV71）和幽门螺杆菌（Hp）的活性。方法：以人肌肉横纹瘤细胞（RD）作为受试细胞,用EV71病毒感染RD细胞,噻唑蓝（MTT）比色法检测裸花紫珠颗粒抗EV71病毒的作用;采用幽门螺杆菌ATCC43504国际标准菌株,将100、50、25、12.5、6.25 mg/mL不同浓度的裸花紫珠颗粒药液进行琼脂打孔法试验,测定不同浓度药物产生的抑菌圈直径大小。结果：裸花紫珠颗粒可抵抗EV71病毒对细胞的感染,治疗指数为35.26;裸花紫珠颗粒对Hp具有一定的抗菌活性,100 mg/mL和50 mg/mL浓度的抑菌圈直径分别为（19.7±0.45） mm和（10.8±0.26） mm。结论：裸花紫珠颗粒有较好的体外抗EV71病毒作用,对防治EV71病毒感染导致的小儿手足口病有一定临床意义;裸花紫珠颗粒对Hp具有良好的抑菌作用,有望用于治疗Hp感染引起的慢性胃炎、胃溃疡。Objective：To study the antiviral activity of Luohua Zizhu Granule in vitro onenterovirus 71（EV71） and Helicobacter pylori（Hp） Methods：Human muscle rhabdomyma cells（RD） were used as test cells. RD cells were transinfected with EV71 virus and the anti-EV71 virus activity of Luohua Zizhu Granule was detected by MTT assay. Hp（ATCC 43504 international standard strain） was used,Luohua Zizhu Granule solutions with the concentrations of 100 mg/mL,50 mg/mL,25 mg/mL,12.5 mg/mL,6.25 mg/mL were prepared,and the antibacterial activity of different concentrations of drugs was determined by the diameters of the inhibition zone in the cylinder plate. Results：Luohua Zizhu Granule could resist the infection by cells EV71 and the therapeutic index was 35.26. It also had certain antibacterial activity against Hp. The diameters of the inhibition zone of Luohua Zizhu Granules solution with concentration of 100 mg/mL and 50 mg/mL were（19.7±0.45） mm and（10.8±0.26） mm,respectively. Conclusion：Luohua Zizhu Granule has good anti-EV71 activity in vitro,which is helpful to prevent hand-foot-mouth disease in children caused by EV71 infection.Antibacterial experiment in vitro showed that Luohua Zizhu Granule has a significant antibacterial effect on Hp. It is promising to use Luohua Zizhu Granule to treat gastric ulcer and chronic gastritis induced by Hp.infection.
