摘 要:目的探讨尾静脉注射法和鼻腔滴注法对肺炎克雷伯菌小鼠模型的影响。方法选择体质量30g左右清洁级雄性昆明小鼠30只,随机分为对照组、尾静脉组、鼻腔组给予菌液后,每2h观察小鼠发病情况,记录小鼠体质量变化。小鼠感染后眼球取血法处死一批小鼠。采用酶联免疫法检测血清C-反应蛋白(CRP)、降钙素原(PCT)和白细胞介素-1(IL-1)、IL-6。苏木精-伊红染色法(HE)染色观察肺组织病理形态学改变。结果对照组小鼠活跃,饮食正常。尾静脉组6h后均开始发病,脸部肿胀,28h死亡1例。小鼠鼻腔滴注细菌后,24h后小鼠逐渐开始发病,36h后小鼠症状加重,对照组小鼠28h和50h体质量为(31.84±0.99)g和(32.31±1.13)g高于感染前(29.93±0.98)g(P<0.001);尾静脉组28h和鼻腔组小鼠28h、50h体质量分别为(26.82±0.52)g和(28.36±0.88)g、(26.06±0.82)g低于对照组相应时期(P<0.001)。尾静脉组小鼠28h体质量低于感染前和鼻腔组28h(P<0.001);随时间延长,鼻腔组体质量逐渐下降(P<0.001)。感染后尾静脉组小鼠CRP、PCT和IL-1、IL-6分别为(44.06±10.37)μg/ml、(18.64±6.92)ng/ml、(158.62±33.86)pg/ml、(19.15±4.82)pg/ml和鼻腔组为(18.39±4.69)μg/ml、(6.23±2.03)ng/ml、(103.03±27.22)pg/ml、(12.18±3.80)pg/ml高于对照组(P<0.001);尾静脉组小鼠CRP、PCT和IL-1、IL-6高于鼻腔组(P<0.001)。尾静脉组小鼠肺组织出现炎性渗出物,肺泡腔内浸润大量炎性细胞,肺泡结构模糊;鼻腔组肺泡出现炎性细胞浸润,肺泡结构呈现模糊。结论尾静脉注射和鼻腔滴注构建肺炎克雷伯菌小鼠模型,体质量、CRP、PCT和炎性因子水平存在差异性,且尾静脉注射比鼻腔滴注严重。OBJECTIVE To investigate the effect of caudal vein injection and nasal drip on Klebsiella pneumoniae model in mice.METHODS A total of 30 clean-grade Kunming mices with the body weight of about 30g were equally divided into the control group,caudal vein group and nasal cavity group.After the bacteria were given,the incidence of pneumonia in the mices was observed every 2 hours and the weight changes of the mices were recorded.After infection in mices,agroup of mice were executed by blood extraction from eyeballs.The serum levels of Creactive protein(CRP),procalcitonin(PCT),interleukin 1(IL-1)and interleukin 6(IL-6)were detected by ELISA.The pathological changes of lung tissue were observed by hematoxylin-eosin staining(HE)staining.RESULTS Mice in the control group were active,with normal diet.The onset of the disease occurred 6 hafter the infection for mice in the tail vein group,swollen face and more eye secretions,and there was 1 case of death in 28 h.The mice instilled with bacteria in the nasal cavity began to show symptoms such as sneezing after 24h,and the symptoms became obviously aggravated after 36 h.The body weight of the control group at 28 hand 50 h was(31.84±0.99)g and(32.31±1.13)g,significantly higher than that before infection,which was(29.93±0.98)g(P〈0.001).The body weight of the caudal vein group and the nasal cavity group at 28 hand 50 hwere(26.82±0.52)g and(28.36±0.88)g,and(26.06±0.82)g,significantly lower than those of the control group at the same time(P〈0.001).The mice of the tail vein group at 28 hweighed significantly less than before infection and the nasal cavity group at the same time(P〈0.001).With the prolonging of time,the body weight of the nasal cavity group decreased significantly(P〈0.001).After infection,the CRP,PCT,IL-1 and IL-6 levels in the caudal vein group were(44.06±10.37)μg/ml,(18.64±6.92)ng/ml,(158.62±33.86)pg/ml,(19.15±4.82)pg/ml,and those in the nasal cavity group were 18.39±4.
