摘 要:目的探讨脑内5-羟色胺(5-HT)含量下降后大鼠前额皮质受体后信号P38 MAPK、PKC和Ca MKⅡ蛋白和基因表达的变化及P2X7受体在其中的作用。方法大鼠分为正常组、对照组、对氯苯丙氨酸(PCPA)组、P2X7受体拮抗剂及激动剂干预组。腹腔注射PCPA使脑内5-HT合成下降,脑室注射P2X7受体拮抗剂和激动剂,采用免疫组织化学、Western blot和实时荧光定量PCR检测前额皮质P38 MAPK、PKC和Ca MKⅡ蛋白和m RNA的表达。结果与对照组比较,PCPA组的P38 MAPK蛋白和m RNA表达增加,而PKC和Ca MKⅡ的蛋白和m RNA表达水平下降。与PCPA组比较,P2X7受体拮抗剂能使P38 MAPK蛋白和基因m RNA表达下降,PKC和Ca MKⅡ的蛋白表达水平升高及m RNA表达水平上升;而P2X7受体激动剂引起P38 MAPK蛋白和m RNA表达水平升高,PKC和Ca MKⅡ的蛋白表达下降和基因m RNA表达水平下降。结论P2X7受体介导了脑内5-HT下降所致的前额皮质受体后信号P38 MAPK、PKC和Ca MKⅡ蛋白和基因表达的变化。Objective To investigate the changes of P38 MAPK, PKC and CaMK II expression and involvement of P2X7 receptors in the prefrontal cortex of rats after cerebral 5-HT depletion. Methods Rats were randomly divided into normal group, sham-operated group (control group), PCPA group, P2X7 receptor antagonistgroup and P2X7 receptor agonist group. 5-HT depletion was induced by intraperitoneal injection of PCPA.Immunohistochemistry, western blot and real-time fluorescence quantitative PCR were performed to analyze the protein and mRNA expression of P38 MAPK, PKC and CaMK II in the prefrontal cortex. Results ExpressionP38 MAPK in the PCPA group increased while PKC and CaMK expression decreased significantly compared with those in control group. Treatment of P2X7 receptor antagonist induced decrease of P38 MAPK expression and increase of PKC and CaMK II expression dramatically when compared with the PCPA group. As supposed, P2X7 receptor agonist worked the opposite way compared with P2X7 receptor antagonist. Conclusions P2X7 receptorsplay an important role in modulating P38 MAPK, PKC and CaMK II expression in the prefrontal cortex in rat modelof cerebral 5-HT depletion.
