摘 要:目的:探究甘氨酸对华法林诱导的瓣膜间质细胞(aortic valve interstitial cells,AVICs)钙化是否有保护作用。方法:提取猪AVICs,通过钙浓度测定、碱性磷酸酶(alkaline phosphatase,ALP)活性测定、茜素红染色等方法检测不同浓度甘氨酸对钙化的作用;real-time PCR探究甘氨酸对成骨分化的作用;Western blot、流式细胞学及TUNEL(Td T-mediated d UTP Nick-End Labeling)染色研究甘氨酸对华法林引起的AVICs凋亡的作用。结果:甘氨酸抑制华法林引起的AVICs钙化,抑制m RNA水平成骨分化标志物的表达,抑制Smad1、5/RUNX2通路,并且抑制华法林引起的AVICs凋亡。结论:甘氨酸可能抑制Smad1、5/RUNX2通路,并且抑制钙化早期凋亡,因此对华法林引起的AVICs钙化具有保护作用。Objective:To investigate if glycine has the protection effect on warfarin induced aortic valve interstitial cells(AVICs)calcification. Methods:Porcine AVICs were isolated,calcium quantification,alkaline phosphatase activation and Alizarin red staining were performed to determine the effect of different concentrations of glycine on calcification,and the m RNA expression of osteogenic markers were determined by RT-PCR. Western blot,Flow cytometry and TUNEL assay were performed to investigate the anti-apoptosis effect on warfarin induced AVICs calcification. Results:Glycine attenuated warfarin induced AVICs calcification,reduced the m RNA expression of osteogenic markers,inhibited the Smad1,5/RUNX2 pathway and decreased the apoptosis of warfarin induced calcification. Conclusion:Glycine inhibits Smad1、5/RUNX2 pathway and decreases the early stage of apoptosis to attenuate warfarin induced AVICs calcification.
