针刺预处理对急性胃黏膜损伤大鼠EGF、TGF-α及TFF3含量的影响

摘　　要：目的观察针刺预处理中脘、足三里穴对急性胃黏膜损伤大鼠胃组织中表皮生长因子(EGF)、转化生长因子-α(TGF-α)及胃黏膜中三叶因子家族-3(TFF3)的影响。方法将32只健康SD大鼠随机分为4组,即空白组、模型组、针刺组、针刺非穴组。针刺预处理8 d采用无水乙醇灌胃的方法制备急性胃黏膜损伤模型。光镜下观察大鼠胃黏膜病理组织形态变化,免疫组化法测定胃黏膜组织中EGF、TGF-α及TFF3的含量。结果与空白组相比,模型组大鼠中胃黏膜损伤指数、EGF、TGF-α及TFF3的表达增强(P<0.01);与模型组、针刺非穴组相比,针刺穴位组TGF-α、EGF的表达升高(P<0.01),而TFF3、胃黏膜损伤指数降低(P<0.01)。结论针刺足三里、中脘穴能够降低无水乙醇对胃黏膜的损伤指数,可通过调节EGF、TGF-α及TFF3的含量,来提高胃黏膜损伤修复的程度,且针刺穴位组对胃黏膜损伤的保护作用具有穴位特异性。Objective To observe the effect of acupuncture pretreatment at Zhongwan and Zusanli points on the epideemal growth taetor （EGF） and transforming growth taetor-alpha （TGF-α） in gastric tissue and the tretbil taetor 3 （TFF3） in gastric mucosa in rats with acute gastric mucosal injury. Methods Thirty-two healthy Sprague-Dawley rats were randomly divided into four groups： blank group, model group, acupuncture group, and non-acupoint acupuncture group. After 8 day-s of acupuncture pretreatment, an acute gastric mucosal injury model was prepared by intragastrie administration of absolute ethanol. The pathologieal ehanges of gastrie mueosa in rats were evaluated by optieal mieroseopy. The levels of EGF, TGF-α, and TFF3 in gastrie mueosa were measured by immunohistoehemistry. Results Compared with the blank group, the model group had signifieantly inereased gastrie mueosal injury index and expression levels of EGF, TGF-α, and TFF3 （P〈 0.01）. Compared with the model group and non-aeupoint aeupuneture group, the aeupoint aeupuneture group had signifieantly higher expression levels of TGF-α and EGF （P〈0.01）, but signifieantly lower level of TFF3 and gastrie mueosal injury index （P〈 0.01）. Conclusion Aeupuneture at Zusanli and Zhongwan points ean reduee the injury index of gastrie mueosa eaused by absolute ethanol. It ean promote the reeovery- from gastrie mueosal injury- by regulating the levels of EGF, TGF-α, and TFF3. The proteetive effeet of aeupuneture against gastrie mueosal injury has aeupoint speeificity.
