摘  要:目的研究唑来膦酸脂质体抑制口腔上皮癌KB细胞增殖的作用。方法用逆向蒸发法制备唑来膦酸脂质体。实验分组为空白组、对照组和实验组。空白组予以新鲜培养基,对照组予以含10μmol·L^(-1)唑来膦酸的完全培养液,实验组予以含10μmol·L^(-1)唑来膦酸脂质体的完全培养液。于干预24,48和72 h后,用噻唑蓝(MTT)比色法考察口腔上皮癌KB细胞的存活率。结果唑来膦酸脂质体的平均粒径为100.72 nm,包封率83.56%,载药量5.21%,平均电位-18.21 mV。干预24 h后,实验组和对照组的存活率分别为(44.87±7.23)%和(65.27±5.11)%;干预48 h后,实验组和对照组的存活率分别为(35.34±4.22)%和(55.11±7.11)%;干预72 h后,实验组和对照组的存活率分别为(22.67±6.54)%和(48.22±8.13)%,差异均有统计学意义(均P<0.001)。结论用逆向蒸发法制得的唑来膦酸脂质体粒径分布均匀,具有较好的稳定性,符合制剂学要求。唑来膦酸脂质体对口腔上皮癌KB细胞的增殖抑制作用明显强于唑来膦酸游离药。Objective To study the inhibitory effect of zoledronic acid liposome on proliferation of oral epithelial carcinoma KB cells. Methods The zoledronic acid liposome was prepared by reverse evaporation. The study was divided into blank,control and test groups. Blank group was given fresh medium. Control group received complete medium containing 10 μmol·L^-1 zoledronic acid. Test group was treated with complete medium containing 10 μmol·L^-1 zoledronic acid liposome. After intervention 24 h and 72 h,the survival rate of oral epithelial carcinoma KB cells was determined by MTT assay. Results The mean particle size was 100. 72 nm,encapsulation rate was 83. 56%,drug-loading rate was 5. 21%,and average potential was -18. 21 mV. In test and control groups,the survival rates after intervention 24 h were( 44. 87 ± 7. 23) % and( 65. 27 ± 5. 11) %,the survival rates after intervention 48 h were( 35. 34 ± 4. 22) % and( 55. 11 ± 7. 11) %,the survival rates after intervention 72 h were( 22. 67 ± 6. 54) % and( 48. 22 ± 8. 13) %,the differences were statistically significant( all P〈0. 001). Conclusion The zoledronic acid liposomes prepared by reverse evaporation can obtain uniform particle size distribution and good stability, meeting therequirements of pharmacology. The proliferation inhibitory effect of zoledronic acid liposome on oral epithelial carcinoma KB cells with was stronger than that of zoledronic acid free drug.
