摘 要:近年,小檗(Berberis)被鉴定是小麦条锈菌的转主寄主,从而发现了小麦条锈菌存在有性生殖,使得研究条锈菌与转主寄主小檗相关的生物学、遗传学成为可能。小麦条锈菌(Puccinia striiformis West.f.sp.tritici Erikss.et Henn.)是活体营养的专性寄生物,只能在活体上侵染繁殖。Puccinia striiformis West. f. sp. tritici Erikss. et Henn. (Pst), is a heteroecious and obligate rust fungus,causing wheat stripe rust in the world. The rust completes sexual cycle on Berberis (mainly) and Mahonia serving as alternate hosts. However, propagation of Berberis plants from seeds to young seedlings is time consuming, hindering rapid identification of Berberis as alternate host for Pst. To know whether Pst completes sexual cycles, including pycnial stage and aecial stage, on Berberis in vitro, we tested each of two stages of sexual cycle Pst on the detached leaves of Berberis (B. shensiana Ahrendt) after the Berberis leaves were inoculated using basidiospores produced from germinated teliospores of Pst race CYR32 in a Petri dish. During cultivation of detached leaves in vitro since inoculation, only water-soaked cotton wrapped around leaf stalk of the leaves. The results showed that Pst performed pycnial and aecial stages on detached leaves of B. shensiana at 9 and 16 d after inoculation in vitro, similar to that of living Berberis plants. The resultant aeciospores were inoculated on wheat cultivar Mingxian 169 and produced a great deal of uredia and urediospores on wheat leaves at 15 d post inoculation, indicating that aeciospores produced on detached leaves of Berberis were vigorous. In vitro, duration of detached leaves of B. shensiana with supply of only water reached up to 35 days, much longer than maximum time length of sexual cycle of Pst completing on living Berberis plants. The present study demonstrated that in vitro test of Berberis susceptibility against Pst using detached young leaves of Berberis was effective and feasible, facilitating testing study of identification of Berberis to serveas alternate host for Pst or genetic study on Pst using Berberis.
