摘  要:目的采用HPLC法同时测定吴茱萸水提物中绿原酸、芦丁、金丝桃苷、去氢吴茱萸碱的含量。方法采用C_(18)色谱柱(100 mm×4. 6 mm,3. 5μm),流动相为乙腈-0. 1%磷酸水混合溶液,梯度洗脱,流速1. 0 mL·min(-1),检测波长326 nm,进样量5μL,柱温30℃。结果绿原酸、芦丁、金丝桃苷、去氢吴茱萸碱的线性范围分别为25. 3~253、0. 501~50. 1、0. 252~25. 2、5. 03~503μg·mL(-1)(r2≥0. 9995),平均加样回收率为100. 0%~100. 9%,RSDs均≤0. 8%。结论所用方法操作简便、结果准确,重复性和稳定性均较好,可作为吴茱萸药材质量控制的方法。OBJECTIVE To simultaneous determine the chlorogenic acid,rutin,hyperoside and dehydroevodiamine in the water extracts of Euodia rutaecarpa. by HPLC. METHODS Separation was performed on an C_(18) column(100 mm × 4. 6 mm,3. 5 μm). The mobile phase consisted of acetonitrile-0. 1% phosphoric acid water with a gradient of 1. 0 mL·min(-1). The detection wavelength was326 nm with the injection volume of 5 μL and the column temperature of 30 ℃. RESULTS The calibration curves of all four constituents showed good linearity( r2≥0. 9995) in a relatively wide concentration ranges. The linear ranges of chlorogenic acid,rutin,hyperoside and dehydroevodiamine were 25. 3-253 μg·mL(-1),0. 501-50. 1 μg·mL(-1),0. 252-25. 2 μg·mL(-1) and 5. 03-503 μg·mL(-1),respectively. The mean recoveries of the four components were 100. 0%-100. 9% with RSDs≤0. 8%. CONCLUSION The method is simple and accurate,and it has excellent repeatability and stability. It can be used as the quality control method for E.rutaecarpa.
