镇肝熄风汤对自发性高血压大鼠STAT1 mRNA和蛋白表达的影响

摘　　要：目的探讨不同剂量的镇肝熄风汤对自发性高血压大鼠(SHR)腹主动脉血管组织STAT1蛋白和STAT1 mRNA表达的影响。方法将60只24周龄雄性SHR大鼠随机分为模型组、中药低剂量组、中药中剂量组、中药高剂量组、复方罗布麻组,每组各12只;将同源雄性魏-凯二氏大鼠(WKY)12只设为正常对照组。灌胃给药45天后,Western Blot测定腹主动脉血管组织中STAT1蛋白表达,实时聚合酶链反应(RT-PCR)方法测定大鼠STAT1 mRNA基因表达。结果与模型组比较,正常组、镇肝熄风汤中药中剂量组和高剂量组STAT1蛋白表达和STAT1 mRNA表达明显降低(P<0.01)。结论镇肝熄风汤可以抑制腹主动脉血管组织中STAT1蛋白表达和STAT1 mRNA表达,可能通过抑制腹主动脉血管细胞凋亡和减少血管细胞增殖的作用,来降低血管重构,从而达到降低血压的作用。Objective To study the effect of different doses of Zhengan Xifeng decoction on STAT1 mRNA and protein expression in the abdominal aortic vascular tissue of spontaneously hypertensive rats（SHR）.Methods 60 male SHR,24 weeks old,were randomly divided into the model,low dose,medium dose,high dose of Zhengan xifeng decoction and the compound apocynum groups.Twelve homologous male rats（WKY）served as the normal control group.After gavage for 45 days,western blotting and RT-PCR were used to detect the STAT1 protein and STAT1 expression in the abdominal aortic vascular tissue.Results Compared with the model group,the STAT1 protein and STAT1 mRNA expression in the normal group,medium dose of herbal medicine group and high dose of herbal medicine group were significantly decreased（F=833.093,sig=0.000,P〈0.01）.Conclusion Zhengan Xifeng Decoction can inhibit the expression of STAT1 protein and STAT1 mRNA in abdominal aorta and may reduce the vascular remodeling by inhibiting the apoptosis of abdominal aorta and reduce the proliferation of vascular cells,which play a role in lowering blood pressure.
