摘  要:厌氧氨氧化菌的储存对于厌氧氨氧化的应用有重要作用。该研究选用以不同质量比例的二甲基亚砜(DMSO)与甘油组成的混合冷冻保护剂,并采用真空冷冻干燥法来获得厌氧氨氧化干菌粉。菌粉在4℃条件下真空遮光保藏75 d。在经过58 d的连续实验复壮后,各复壮反应器的比厌氧氨氧化活性(SAA)与污泥特性显示出不同的特点。实验结果说明,以1.5%(质量分数)DMSO+1.5%(质量分数)甘油组成的混合保护剂保护的厌氧氨氧化污泥的复壮效果最好,在运行稳定时期其平均SAA为85.5 mg N/(g VSS·d),活性恢复率为65.8%,其厌氧氨氧化化学计量比(Rs与Rp)分别为1.56与0.57,胞外聚合物(EPS)含量为31.15 mg/g VSS,而蛋白质/多糖(PN/PS)为0.96。The preservation of anaerobic ammonium oxidation(anammox) bacteria is crucial to application of the anammox process. In this experimental study, a mixed cryoprotectant consisting of different mass proportions of dimethylsulfoxide(DMSO) and glycerol was prepared to obtain dry powder of anammox by use of the vacuum freeze-drying method, and the powder was stored under the condition of vacuum, dark and temperature 4 ℃ for 75 days. After 56 days of continuous rejuvenation experiment, it was showed that characteristics of the SAA(specific anaerobic ammonia oxidation activity) and anaerobic sludge were different in each reactor. The experiment suggested that the rejuvenation effect of anammox sludge being protected by a mixture of 1.5%(mass fraction) DMSO + 1.5%(mass fraction) glycerol was the best: during the stable operation period, the average SAA was 85.5 mg N/(g VSS·d), the activity recovery rate was 65.8%, the anammox stoichiometric ratio(Rs, Rp) being respectively 1.56 and 0.57, the extra-cellular polymer(EPS) content was 31.15 mg/g VSS and the ratio of protein/polysaccharide(PN/PS) 0.96.
