摘  要:目的探讨白簕多糖的提取工艺,研究白簕多糖分散片的制备工艺。方法用水提醇沉法提取白簕多糖,采用正交试验优选提取时间和次数;单因素考察醇沉量,优化提取工艺;单因素考察并确定分散片处方中的辅料和用量。结果白簕多糖最佳提取工艺为分别加12倍和10倍水各提取2 h,浓缩后加1.5倍70%乙醇醇沉。白簕多糖干膏粉:磷酸氢钙为1:2,水为湿润剂,二氧化硅为助流剂,交联聚维酮(PVPP)为崩解剂(内加2%,外加5%),0.5%硬脂酸镁为润滑剂制得白簕多糖分散片每片40 mg。多糖含量稳定,溶出度大于85%,收率稳定,适合生产的要求。结论优化白簕多糖提取工艺,分散片制备工艺稳定,可用于生产。Objective To study extracting technology of acanthopanax trifoliatus polysaccharide,and to research preparation of dispersible tablets of acanthopanax trifoliatus polysaccharide. Methods The method of water extracting-alcohol precipitating was used to extract acanthopanax trifoliatus polysaccharide, optimized extraction time and times by orthogonal selection. The amount of alcohol precipitation by single factor exploration, optimized the extracting technology. Determined the excipients and amount of the dispersible tablets by single factor exploration. Results The optimum extraction technology of acanthopanax trifoliatus polysaccharide were 12 times and 10 times the water extraction for 2 hours respectively. After concentrated, the ethanol precipitation was 1.5 times of 70% ethanol. Dry extract powder of acanthopanax trifoliatus polysaccharide and calcium hydrogen phosphate at the ratio of 1:2. Water as wetting agent, Silica as flow aid, PVPP as disintegrant (internal addition by 2%,External addition by 5%), magnesium stearate as lubricant, prepared dispersible tablets of acanthopanax trifoliatus polysaccharide, 40 mg/ tablet. The content of polysaccharide was stable, the dissolution rate was more than 85%, the yield was stable, it was suitable for production requirements. Conclusion The extracting technology of acanthopanax trifoliatus polysacchadde is optimized, the preparation of dispersible tablets are stable, and can be used in pilot scale production.
