摘  要:为确定适宜于加工番茄花药愈伤组织诱导的最佳取材时期和激素组合,以2个加工番茄品种的花药为材料,以醋酸洋红液为染液,压碎法制片,用体视显微镜观察小孢子的发育时期;以MSB培养基为基本培养基,附加不同浓度的植物激素2,4-D、NAA、KT、6-BA、ZT、TDZ进行花药培养,研究不同植物激素组合对加工番茄花药愈伤组织诱导的影响。结果表明:番茄花蕾纵径为5.25-6.19 mm,横径为2.09-3.20 mm,绝大多数的小孢子发育时期处于单核靠边期;不同基因型加工番茄愈伤组织的诱导率不同,品种CK8的诱导率为18.63%,品种CK3的诱导率为14.19%;0.5 mg/L 2,4-D+0.05 mg/L TDZ是最适宜愈伤组织诱导的激素组合,诱导率为56.52%,其次是0.1 mg/L2,4-D+2.0 mg/L 6-BA的组合,诱导率为56.36%;番茄花药愈伤组织在MS+0.2 mg/L NAA+0.5 mg/LKT培养基上可形成胚性愈伤组织;4℃低温预处理花蕾的适宜时间为2-5 d。The aim of this study is to determine the suitable stages and hormones combinations for inducing callus of the processing tomato, and anthers of two varieties processing tomato were used as materials. Anthers were crushed and made tablets with acetic acid magenta, and then the spore development stage was observed by stereomicroscope. MSB medium supplemented with different concentrations of plant hormones such as 2,4-D, NAA, KT, 6-BA, ZT and TDZ for anther culture were used to study the effects of different combinations on callus induction of processing tomato anthers. The results showed that the longitudinal diameter of tomato flower buds was 5.25-6.19 mm and the transverse diameter was 2.09-3.20 mm, and most of the microspores were in the mononuclear development stage. The callus induction rates were varied in different tomato genotypes, and the induction rates were 18.63% and 14.19% in CK8 and CK3, respectively. The combination of 0.5 mg/L 2,4-D with 0.05 mg/L TDZ was the most suitable hormone combination for callus induction, the induction rate was 56.52%, followed by the combination of 0.1 mg/L 2,4-D and 2.0 mg/L 6-BA, and the induction rate was 56.36%. The embryogenic calluses were formed on MSB supplemented with 0.2 mg/L NAA and 0.5 mg/L KT. Suitable time for low temperature pretreatment of buds at 4 ℃ was 2-5 d.
