摘  要:云南地方桃资源丰富,冲天桃为其中一种,为了寻找云南省地方桃资源中具有高抗病的种质资源,从而为云南省抗病性地方桃品种选育提供理论基础,本研究根据已知植物抗病基因的NBS-LRR类保守序列设计简并引物,以冲天桃为试材,对其基因组DNA进行PCR扩增、克隆、测序,共得到5条NBS类抗病基因同源序列(RGAs),大小约为500 bp,经序列分析表明:这5条RGAs与桃亚属植物中推测的一些抗病基因蛋白具有较高的同源性,其中一些RGAs与番茄抗青枯病基因Prf及烟草的抗花叶病毒N基因具有高度的同源性。因此可以确定‘冲天桃’植株中存在较为丰富的NBS-LRR类抗病基因,这为下一步云南省抗病性地方桃品种选育提供了研究依据。There are rich local peach resources in Yurman, and 'chongtian peach' is one of them. In order to find the germplasm resources with high resistance to disease in the local peach resources in Yunnan Province and provide a theoretical basis for the breeding of disease-resistant local peach varieties in Ytmnan Province, in this study, the design of degenerate primers was based on the NBS-LRR class conservative sequence of known plant resistance genes. Taking 'chongtian peach' as a test material, PCR amplification, cloning and sequencing of its genomic DNA were conducted. A total of 5 NBS class resistant gene analogs (RGAs) were obtained, and the size was about 500 bp. Sequence analysis showed that these 5 RGAs were highly homologous to some of the disease-resistant proteins in the plants of peach subgenus. Some of the RGAs genes were highly homologous to the gene Prf against bacterial wilt of tomato and the N gene of tobacco mosaic virus. Therefore, it could be confirmed that there were abundent NBS-LRR class resistance genes in the plants of Prunus persica, which might lay the foundation for further breeding of disease-resistant local peach varieties in Yunnan Province.
