摘  要:表皮蜡质层和角质膜对植物自我防护外界生物和非生物胁迫发挥重要调控作用,β-酮脂酰-CoA合酶(KCS)基因是表皮蜡质层中超长链脂肪酸合成反应中的限速酶,为揭示β-酮脂酰-CoA合酶与植物抗旱特性之间的关系,本研究从黄麻中分离了β-酮脂酰-CoA合酶(KCS)基因,该基因cDNA全长1 572 bp,编码523个氨基酸,将该基因正向插入植物表达载体(pCAMBIA2300)中,利用花絮侵染法转化拟南芥,通过PCR和Southern blotting分子检测表明,外源基因已整合到拟南芥基因组中,模拟干旱胁迫结果表明,过表达黄麻β-酮脂酰-Co A合酶基因的拟南芥与野生型相比,转化植物的根根毛数量、根长及抗旱性能有不同程度的提高。该研究结果为进一步解析β-酮脂酰-CoA合酶基因参与调控植物抗旱的分子机制提供指导。Cuticular wax layer and cuticular membrane stress play an important role in the regulation of plant selfprotection against the biotic and abiotic. β-ketoacyl-CoA synthase (KCS) gene was the rate limiting enzyme of acid synthesis process in super long chain fatty cuticular waxes. In order to uncover the function of β-ketoacyl CoA synthase gene in plant drought resistance response, we isolated β-ketoacyl-CoA synthase (KCS) gene from jute, the cDNA contained 1 572 bp, encoded 523 amino acids. And we took this gene forward insertion plant expression vector (pCAMBIA2300), using dipping flower method transformed into A rabidopsis, and PCR and Southern blot showed that the exogenous gene has been integrated into the genome of A rabidopsis thaliana. The results of drought dress showed that A rabidopsis of the over-expression of jute β-ketoacyl-CoA synthase gene compared with the wild type, the transgenic plants had longer root, more root hairs and more drought tolerant performance. The results of this study might lay the foundation for analyzing the molecular mechanism of regulating by β-ketoyl-CoA synthase gene in plant drought resistance.
