摘  要:目的观察七氟烷后处理对脑缺血-再灌注大鼠脑组织toll样受体4(TLR4)、肿瘤坏死因子受体相关因子6(TRAF6)表达的影响。方法选取健康SD雄性大鼠随机分为假手术组(C组)、缺血-再灌注组(I-R组)、1. 0MAC七氟烷后处理组(S1组)、1. 5 MAC七氟烷后处理组(S2组),根据再灌注时间分为6 h、12 h、24 h、48 h亚组(n=10)。采用线栓法制备缺血-再灌注模型。C组除不插线栓进行缺血操作外,其余步骤同脑缺血-再灌注建模操作。I-R组行脑缺血2 h,并根据分组进行持续再灌注6 h、12 h、24 h、48 h。S1、S2组在I-R组的基础上进行七氟烷后处理。对4组大鼠进行神经功能缺损评分,TCC染色测定脑梗死体积,免疫组化法检测脑组织TLR4、TRAF6蛋白表达水平,荧光定量PCR检测TLR4、TRAF6 mRNA表达,TUNEL染色检测细胞凋亡。结果与C组各亚组比较,I-R组可见明显神经功能缺损、缺血侧大脑半球梗死、脑组织细胞凋亡,七氟烷后处理大鼠神经功能缺损评分显著降低、大脑梗死体积显著减小、细胞凋亡指数(AI)显著降低(P <0. 05),且S2组显著优于S1组(P <0. 05);组内比较,再灌注24 h组脑梗死体积、细胞凋亡指数(AI)显著高于其他亚组(P <0. 05)。与C组各亚组比较,I-R组相应亚组TLR4、TRAF6蛋白及mRNA表达显著升高(P <0. 05),七氟烷处理后有所降低(P <0. 05),且S2组显著低于S1组(P <0. 05);组内比较,再灌注后24 h组TLR4、TRAF6蛋白及mRNA表达显著高于其他亚组(P <0. 05)。结论七氟烷后处理可减轻缺血-再灌注大鼠脑组织损伤及神经功能缺损,降低脑组织TLR4及TRAF6的表达。Objective To investigate the effects of seven sevoflurane postcondionting on the expression of toll-like receptor 4 (TLR4) and tumor necrosis factor receptor-associated factor 6 (TRAF6) in brain tissue of rats after cerebral ischemia-reperfusion. Methods Healthy male SD rats were randomly divided into sham operation group (C group), ischemia-reperfusion group (I-R group), 1.0MAC group of seven sevoflurane postprocessing (S1 group), 1.5MAC group of seven sevoflurane postprocessing (group S2), and each group was divided into 6 h, 12 h, 24 h, and 48 h subgroup (n=10) according to the time of reperfusion. The cerebral ischemia-reperfusion model was established using Zea Longa suture method. The neurological function was evaluated in rats, the infarction volume was measured by TCC staining, the TLR4 and TRAF6 protein expression levels in brain tissue were detected by immunohistochemical method, the TLR4 and TRAF6 mRNA expression levels in brain tissue were detected by fluorescence quantitative PCR , and apoptosis condition in brain tissue was detected using TUNEL. Results Compared to each subgroup of C group, I-R group showed obvious neurologic impairment, cerebral hemisphere infarction and brain cell apoptosis, after seven sevoflurane postcondionting, neurological deficit scores of rats was significantly decreased, the brain infarct volume was significantly decreased and the apoptosis index (AI) was decreased significantly ( P 〈0.05), and group S2 was significantly better than group S1 ( P 〈0.05). Compared with the other subgroups, the volume of cerebral infarction and the apoptotic index (AI) of the 24 h group were significantly higher ( P 〈0.05). Compared to each subgroup of C group, protein and mRNA expression of TLR4 TRAF6 in I-R group were significantly increased ( P 〈0.05), after seven sevoflurane postcondionting, protein and mRNA expression of TLR4 and TRAF6 in brain tissue were decreased ( P 〈0.05), and group S2 was significantly lower than
