摘 要:目的观察银杏叶提取物对老年大鼠肝叶部分切除术后认知功能障碍及大脑海马CA3区凋亡影响。方法雄性健康SD老年大鼠60只,随机数字表法分为三组:对照组(C组)、模型组(M组)和银杏叶提取物治疗组(T组),每组20只。C组未进行任何处理,M组行肝叶部分切除术建立大鼠术后认知功能障碍模型,T组术前连续5 d每天腹腔注射100 mg/kg银杏叶提取物+行肝叶部分切除术建立大鼠术后认知功能障碍模型。通过Morris水迷宫实验测定大鼠逃避潜伏期、原平台探索时间及穿越平台次数,通过Tunnel法检测大鼠CA3区凋亡情况,通过Western blot法检测海马大鼠CA3区Bcl-2、Bax表达情况。结果相比于C组,M组大鼠术后第1、2、3、5、7天逃避潜伏期延长,平台象限探索时间缩短,穿越平台次数减少,海马CA3区凋亡增加,Bax表达上调,Bcl-2表达下调,Bcl-2/Bax值降低(P <0. 05)。相比于M组,T组大鼠术后第3、5、7天逃避潜伏期缩短,平台象限探索时间延长,穿越平台次数增加,海马CA3区凋亡减少,Bax表达下调,Bcl-2表达上调,Bcl-2/Bax值升高(P <0. 05)。结论银杏叶提取物可抑制海马细胞凋亡,从而改善大鼠术后认知功能障碍。Objective To explore the effect of ginkgo biloba extract on postoperative cognitive dysfunction and apoptosis of hippocampal CA3 region in aged rats. Methods 60 male healthy SD rats were randomly divided into three groups: the control group (C group), model group (M group) and ginkgo biloba extract treatment group (T group)(n=20). The C Group did not receive any treatment, M Group underwent partial hepatectomy to establish a rat model of postoperative cognitive impairment, T Group received intraperitoneal injection of 100 mg/kg Ginkgo biloba extract for 5 consecutive days, then, underwent partial hepatectomy to establish a rat model of postoperative cognitive impairment. The Morris water maze experiment was used to determine rats escape latency, original platform exploration time and frequency of crossing the platform, the Tunnel method was used to detect apoptosis level of hippocampal CA3 region, the Western blot was used to detect the expression of Bcl-2 and Bax. Results Compared with C group, the escape latency was prolonged, original platform exploration time was shortened, frequency of crossing the platform was reduced in M group at 1th, 2th, 3th, 5th, 7th day after surgery ( P 〈0.05). In addition, compared with C group, the apoptosis was increased in hippocampal CA3 region, the expression of Bax was increased, Bcl-2 was decreased, the ratio of Bcl-2/Bax was decreased in M group ( P 〈0.05). Compared with M group, the escape latency was shortened, original platform exploration time was prolonged, frequency of crossing the platform was increased in T group at 3th, 5th, 7th day after surgery ( P 〈0.05). In addition, compared with M group, the apoptosis was reduced in hippocampal CA3 region, the expression of Bax was decreased, Bcl-2 was increased, the ratio of Bcl-2/Bax was increased in T group ( P 〈0.05). Conclusion Ginkgo biloba extract can inhibit the apoptosis of hippocampal cells and improve the postoperative cognitive dysfunction.
