摘 要:目的探究六味安消胶囊对慢传输型便秘(STC)大鼠肠道功能、排便量、肠神经递质及水通道蛋白(AQP)的影响。方法雄性SD大鼠60只,随机分为对照组、模型组、六味安消胶囊组(30 mg/kg,40 mg/kg,50 mg/kg)及莫沙必利组;模型组、六味安消胶囊组及莫沙必利组采用复方地芬诺酯混悬液10 mg/(kg·d)灌胃,连续给药14 d,建立STC模型;模型建立后,六味安消胶囊组及莫沙必利组分别给予六味安消胶囊(30 mg·kg^(-1),40mg·kg^(-1),50 mg·kg^(-1))及莫沙必利混悬液1.5 mg/(kg·d)灌胃,对照组及模型组给予等体积0.9%Na Cl溶液灌胃,连续给药14 d。检测各组大鼠造模前、造模期及治疗期粪便数量及含水量;计算各组大鼠碳末推进率;蛋白免疫印迹(Western blot)检测各组大鼠结肠组织水通道蛋白1,3,4,8(AQP1,3,4,8)、干细胞因子及其受体(SCF/c-kit)蛋白表达。结果与对照组相比,模型组大鼠碳末推进率、排便量及粪便含水量降低,模型组大鼠结肠AQP1、3、4、8表达升高,SCF、c-kit蛋白表达降低(P<0.05);与模型组相比,六位安消胶囊组及莫沙必利组大鼠碳末推进率、排便量及粪便含水量升高,结肠AQP1,3,4,8表达降低,SCF/c-kit蛋白表达升高(P<0.05)。结论六味安消胶囊可改善STC大鼠肠道功能、排便数量及粪便含水量,其机制可能与改善AQPs蛋白及SCF/c-kit信号通路有关。Objective To explore the effect of liuwei anxiao capsule on intestinal function, defecation, intestinal neurotransmitter and aquaporin (AQP) in rats with slow transit constipation (STC). Methods 60 male SD rats were randomly divided into the control group, the model group, the liuwei anxiao capsule group (30 mg/kg, 40 mg/kg, 50 mg/kg), and the moxa bili group. In the model group, the liuwei anxiao capsule group and the moxa bili group, the compound was used to fill the stomach of 10 mg/(kg·d), and the STC model was established by continuous administration of 14 d. After the model was established, the liuwei anxiao capsule group and the mosha bili group were respectively given the liuwei anxiao capsule (30 mg/kg, 40 mg/kg, 50 mg/kg), and the moxapine suspension of 1.5 mg/(kg·d). The control group and the model group were given 0.9% NaCl solution to fill the stomach and continuously given the drug 14 d. The amount of feces and water content of the rats were examined before and after the models were made. The carbon end rate of rats in each group was calculated. Western blot was used to detect the expression of water channel protein 1, 3, 4, 8 (AQP1, 3, 4, 8) and SCF/c-kit protein in rats of rats. Results Compared with control group, the end of the carbon propulsion rate, amount of defecation and fecal water content of model group rats were decreased, colon AQP1, 3, 4, 8 of the model group rats were in higher expression, SCF, c - kit protein expression was reduced ( P 〈0.05). Compared with the model group, the carb propelling rate, defecation volume and fecal water content of the rats in the liuwei anxiao capsule group and moxa bili group were increased, and the expression of AQP1, 3, 4, 8 in the colon was lower than that in the model group. The expression of SCF/c-kit protein was increased ( P 〈0.05).
