摘  要:目的探讨转化生长因子β1(TGFβ1)参与的脂肪干细胞-富血小板血浆凝胶促进大鼠创面修复的机制。方法 80只清洁级SD大鼠随机分为5组:对照组、模型组、脂肪干细胞组、富血小板凝胶组和脂肪干细胞联合富血小板凝胶组(n=16)。除对照组大鼠外,其余各组大鼠在背部制作4个1 cm^2的正方形皮肤全层创面缺损创面,脂肪干细胞组、富血小板凝胶组和脂肪干细胞联合富血小板凝胶组大鼠分别给予对应的药物处理。分别于给药处理3 d、5 d、7d和14 d后处死动物,观察各组大鼠的创面愈合情况及创面组织中羟脯氨酸、TGFβ1-Smad蛋白的表达。结果创面损伤大鼠给药3 d后富血小板凝胶组和脂肪干细胞联合富血小板凝胶组大鼠创面比模型组和脂肪肝细胞组干燥,给药14 d后各组大鼠创面基本恢复,效果具有时间依赖性(P <0. 05);创面损伤大鼠创面组织中羟脯氨酸、TGFβ1、Smad1/3及MMP2/9蛋白水平明显低于对照组(P <0. 05),而经脂肪干细胞、富血小板凝胶、脂肪干细胞联合富血小板凝胶治疗后上述蛋白水平明显升高(P <0. 05),且脂肪干细胞联合富血小板凝胶治疗组和给药14 d后效果最明显(P <0. 05),效果具有时间依赖性(P <0. 05)。结论脂肪干细胞-富血小板血浆凝胶具有显著促进大鼠创面修复的效果,其可能与调控TGFβ1-Smad及基质金属蛋白酶表达有关。Objective The present research tried to explore the role of TGFβ1 in repairing soft tissue wounds by platelet rich plasma gel combined with adipose derived stem cells in rats. Methods 80 Clean SD rats were randomized into 5 groups, namely, control, model, platelet rich plasma gel group, adipose derived stem cells group and combination group ( n - 16). Four 1 cm2 wounded skin squares were prepared except the control. All the prepared rats were treated with corresponding compounds for 14 d. At 3,5,7, 14 d, rats were sacrificed for wounds observation and Hyp and TGFβ1 - Smad assays. Results Nearly all wounds recovered 14 d posttreatment, manifesting a time dependent manner of recovery ( P 〈 0.05). The expressions of Hyp, TGFβ1, Smad1/3 and MMP2/9 were lower in wounded rats but rebounded greatly posttreatment ( P 〈 0.05 ) , with the combination group topping the three wounded groups ( P 〈 0.05). Conclusion Platelet rich plasma gel combining adipose derived stem cells might greatly promote the recovery of wounded skin in rats through regulation of TGFβ1- Smad and MMPs.
