摘 要:目的探讨脊髓星形胶质细胞connexin 43(CX43)调控CCL2释放在结直肠扩张致慢性内脏痛中的作用。方法新生期SD大鼠,出生一天随机分成两组,慢性内脏痛(CRD)组及正常组。CRD组建立CRD模型,正常组大鼠除未进行结直肠扩张外,其他操作均与模型组一致。常规饲养至8周成年采用腹壁撤离反射(AWR)评分及痛阈评价内脏痛觉敏感性。鞘内注射CX43拮抗剂、CCR2拮抗剂后检测痛阈。Western blotting检测CX43的表达,ELISA检测CCL2表达。结果与对照组相比,CRD组在20 mmHg、40 mmHg、60 mmHg、80 mmHg压力下AWR明显增加(P <0. 05),痛阈均明显下降(P <0. 05)。鞘内注射CX43拮抗剂后痛阈升高(P <0. 05),CX43、CCL2表达量减少(P <0. 05);鞘内注射CCR2拮抗剂后痛阈升高(P <0. 05)。结论 CRD模型大鼠均存在内脏痛敏,这可能与脊髓星形胶质细胞CX43表达增多进而通过其形成的缝隙连接及半通道释放的CCL2增多有关。Objective To investigate the role of spinal astrocyte connexin 43 ( CX43 ) in regulating CCL2 release in rats with chronic visceral pain caused by eoloreetal dilatation. Methods Newborn SD rats were randonfized into two groups, the normal group and the eoloreetal distention (CRD) group which underwent eoloreetal dilatation. Visceral pain sensitivity was assessed regarding abdonfinal withdraw reflex (AWR) score and pain threshold, which was measured after intratheeal injection of CX43 antagonist and CCR2 antagonist. The expressions of CX43 and CCL2 were analyzed by Western blot and ELISA respectively. Results The AWR in CRD group significantly increased at 20 mmHg, 40 mmHg, 60 mmHg, and 80 mmHg compared with normal ( P 〈 0.05) with the pain threshold significantly decreased ( P 〈 0.05). After intrathecal injection of CX43 antagonist, the pain threshold increased ( P 〈 0.05) , and the expression of CX43 and CCL2 decreased ( P 〈 0.05 ). The pain threshold increased after intratheeal injection of CCR2 antagonist ( P 〈0.05). Conclusion The visceral allodynia in CRD model rats nfight be related to the increased expression of CX43 in spinal astroeytes which regulates release of CCL2 through gap junction and henri - channel.
