摘 要:一直以来,大段骨缺损的修复是骨科临床的难题之一。采用组织工程学方法,构建搭载成血管潜能种子细胞的组织工程骨可以实现材料内部早期血管化,为成功修复大段骨缺损提供了新思路。目前,具有成血管潜能的种子细胞来源多样,各具优势。共培养和预处理技术的应用实现了细胞-细胞、细胞-微环境之间有效的信息交流,定向诱导种子细胞向内皮细胞分化;运用基因编辑及基因转染技术对种子细胞进行特定改造,使其精确表达促血管生长因子,达到"1+1> 2"的效果。同时,种子细胞在定向诱导的过程中存在来源有限、耗时较长、目的蛋白表达低下等不足,细胞-细胞、细胞-微环境之间作用的具体分子学机制尚不清楚,经特定基因转染后的种子细胞的安全性及有效性缺乏长期观察,仍有待于今后深入探索。The repairment of large segment bone defect has always been one of the challenges in orthopedic clinic. Tissue engineering method is used to construct tissue engineering bone with vascular potential seed cells which can realize early vascularization within the material and provides a new idea for successful repair of this kind of bone defect. At present, the seed cells with vascular potential have various sources and advantages ; The application of co-culture and pretreatment technology- realizes the effective exchange of ilfformation between cell-cell and cell-nficroenvironment, which can induce the differentiation of seed ceils to endothelial cells;The gene editing and gene transfection technology- were used to modify the seed cells in order to express exactly the vascular growth factor and achieve the effect of " 1 + 1 〉2". At the same time, in the process of directional induction, for seed cells there are some problems, such as linfited source, time consunfing, low expression of interest protein,etc. The specific molecular mechanisnl of cell-cell, cell-nficroenvironment interaction remains unclear. The safety and effectiveness of seed cells transfected with specific genes are not observed over a long period of time. All these problems remain to be explored more deeply in the future.
