摘 要:目的探究氨基羟乙酸(AOAA)对慢性酒精中毒大鼠学习记忆能力、海马硫化氢、线粒体形态结构、线粒体ATP酶活性和纤维状肌动蛋白(F-actin)含量的影响。方法将60只SD雄性大鼠随机平均分为3组,除正常对照(NC)组外,每组饮含6%(V/V)酒精水溶液28 d,酒精溶液每日9∶00配制,置换。14 d后,AOAA(剂量为5 mg/kg)溶解在1 ml生理盐水中,AOAA治疗(AR)组每日腹腔注射一次,连续14 d。NC组每日腹腔注射1 ml生理盐水。结束后采用Y型电迷宫测试大鼠的学习记忆能力,随后检测海马组织H2S含量、ATP酶活性及F-actin表达,并在电镜下观察海马组织细胞内线粒体形态结构。结果与NC组相比,模型(M)组学会所需训练次数和H2S含量均升高,线粒体ATP酶活性和海马组织区F-actin阳性表达均降低,有极显著性差异(P<0. 01);与M组相比,AR组学会所需训练次数和H2S含量均降低,ATP酶活性和海马组织F-actin阳性表达均升高,有极显著性差异(P<0. 01)。结论 AOAA能够减轻慢性酒精中毒大鼠的症状,改善学习记忆能力,可能与AOAA影响H2S的含量、线粒体结构、Factin的含量有关。Objective To explore the effects of aminooxyacetic acid( AOAA) on the learning and memory abilities,the content of hydrogen sulfide( H2 S),morphology and structure of mitochondria,mitochondrial ATP enzyme activity and the expression of fibrous actin( Factin) in hippocampus of rats with chronic alcoholism.Methods 60 SD male rats were randomly divided into three groups on average.Except for the control group,the rats of each group were fed with the water containing( V/V) 6% alcohol for 28 d. The rats in treatment group were injected AOAA intraperitoneally once a day for 14 d. The rats in normal control group were injected with 1 ml normal saline intraperitoneally once a day.After that,the learning and memory abilities were tested by Y-shaped maze detection.Subsequently,the content of H2 S,mitochondrial ATP enzyme activity and the expression of F-actin in hippocampus of the rats were measured and the morphology and structure of mitochondria in hippocampus were observed by electron microscope. Results Compared with the training time for requiring( 40. 250 0 ±4.008 3),the content of H2 S( 27.430 5±1.520 4),mitochondrial ATP enzyme activity( 2.096 8±0.033 9) and the positive expression of Factin in hippocampus( 0.056 3±0.002 6) of rats in control group,the training times for requiring( 83.562 5±4.925 7),the content of H2 S( 44.505 6±2.401 3) of model group were increased,the mitochondrial ATP enzyme activity( 1.485 3±0.042 9) and the positive expression of F-actin in hippocampus( 0.031 1±0.002 0) of model group were decreased.Compared with those of model group,the training time for requiring( 64.437 5±3.915 2),the content of H2 S( 36.707 5±2.123 3) of treatment group were decreased,the mitochondrial ATP enzyme activity( 1.631 2±0.053 7) and the expression of F-actin in hippocampus( 0.039 9±0.002 3) of treatment group were increased( P〈0. 01).Conclusions AOAA could ease the symptom and improve the abilities of learning and memory in chroni
