摘  要:目的研究持续性心房颤动(房颤)形成过程中,心房肌超极化激活环核苷酸门控(HCN) 2和HCN4通道的变化。方法 12只家犬分为两组,其中持续性房颤模型犬6只(实验组,通过使用右心房快速起搏8 w获得),窦性心律模型犬6只(对照组)。采用实时荧光定量聚合酶链反应和蛋白免疫印迹的方法检测左心房肌HCN2通道和HCN4通道的mRNA和蛋白的表达含量。结果与对照组相比,实验组左心房肌HCN2和HCN4 mRNA表达水平呈明显增高的趋势(P<0. 05),并且其相应蛋白质表达含量也呈明显增高趋势(P<0. 05)。结论持续性心房颤动模型犬左心房肌HCN2和HCN4表达增高,提示HCN2和HCN4通道参与持续性房颤心房电重构。Objective To study the changes of HCN2 and HCN4 channels in the atrial myocyte during the process of persistent AF.Methods 12 dogs were divided into two groups,6 dogs in persistent atrial fibrillation model( right atrial rapid pacing for 8 weeks,continuous atrial fibrillation),and 6 sinus rhythm dogs. The expressions of mRNA and protein in the HCN2 channel and HCN4 channel of the left atrial muscle were detected by real time fluorescence quantitative polymerase chain reaction and immunoblotting. Results Compared with those of sinus rhythm model group,the expression levels of HCN2 and HCN4 mRNA in left atrial muscle of persistent atrial fibrillation group were increased significantly( P〈0.05),and the expressions of corresponding protein were increased significantly( P〈0.05).Conclusions The expressions of HCN2 and HCN4 in the left atrial muscle of persistent atrial fibrillation model dogs are higher,suggesting that HCN2 and HCN4 channels are involved in atrial electrical remodeling of persistent atrial fibrillation.
