摘  要:目的研究大鼠脑震荡伤后主要外周器官HSP70、TNF-α表达量的变化规律,并结合二者比值来推断脑震荡的损伤时间。方法制作大鼠脑震荡模型后,取不同时间点的心、肝、脾、肺、肾组织进行HSP70和TNF-α免疫组织化学染色,分析图像获得二者各自表达量及比值的变化规律,并进行统计学处理。结果 (1)脑震荡后0.5h至12h时间段内,TNF-α及HSP70的表达量均呈现同时上升的趋势,但HSP70表达量均大于TNF-α,其二者表达量之比由1.19下降至本时间段内最低值0.76,提示当肝脏中HSP70、TNF-α的表达量之比在1.19以内,且HSP70的表达量大于TNF-α,则脑震荡损伤时间应在12h以内;(2)心、脾、肺、肾组织中二者的变化规律与肝脏类似。结论观察脑震荡后肝脏、心脏、脾脏、肾脏组织中HSP70、TNF-α的表达量,并结合二者比值的变化情况进行分析,可在一定程度上推断脑震荡的损伤时间范围。按照该研究思路进一步深入研究,有望提高脑震荡损伤时间范围推断的准确度。Objective To study the expression changing trend of HSP70 and TNF-α in in the major peripheral organs of brain concussion injury in rats, and to combine these two indicators' ratio to determinate the post-injury time of brain concussion. Method After the rat brain concussion model was established, tissues were taken from heart, liver, spleen, lung and kidney at different time points, then they were immunohistochemically stained, the resulting images were analysed to reveal the relationship between the expression of HSP70, TNF-α as well as their ratio, and the statistical analysis was processed. Result (1)In the period from 0.5h to 12h after the occurrence of the brain concussion, the expressions of HSP70 and TNF-α both showed an increasing trend, but the expression of HSP70 was always higher than that of TNF-α.At the same time, the ratio of these two indicators in liver goes down from 1.19 to the lowest value 0.76. This consequence prompts us that when the HSP70 and TNF-α expression ratio ≤1.19, and the HSP70 positive cells 〉 TNF-α the post-injury time would be within 12 hours. (2) The situations in heart, spleen, lung and kidney were similar to liver. Conclusion To observe the expressions of HSP70 and TNF-α in liver, heart, spleen, lung and kidney in brain concussion individuals, combining these' ratio and to analyze it, it could be used to infer the time range of brain concussion injury. Based on this idea, a further research is likely to improve the accuracy of brain concussion injury time range inferring.
