摘　　要：目的比较GlobalFiler、PowerPlex 21及Identifiler Plus3种PCR扩增试剂盒对陈旧口腔拭子的检验效率。方法采用硅珠法提取150份保存7-13年陈旧口腔拭子的DNA,分别用GlobalFiler、PowerPlex 21及Identifiler Plus试剂盒进行扩增检测,比较分析所得分型数据。结果在150份降解样本的检验结果中,GlobalFiler试剂盒21个常染色体STR基因座有效分型检出率为76%,PowerPlex 21试剂盒20个STR基因座检出率为74.93%,Identifiler Plus所含的15个STR基因座检出率为77.56%。结论对于陈旧口腔拭子检材,Identifiler Plus检验成功率最高,需要更多基因座时,GlobalFiler优于PowerPlex 21。Objective To compare the performances of GlobalFiler kit, PowerPlex 21 kit and Identifiler Plus kit in aged buccal swabs profling. Methods DNA were extracted from 150 aged buccal swabs by silica-beadsmethod. The DNA were amplified by the GlobalFiler, PowerPlex 21 and Identifiler Plus separately and profiled through the GeneMapper ID-X. Results 76%of 21 autosomal loci were successfully typed by GlobalFiler kit in degraded samples, 74.93% of 20 autosomal loci were successfully typed by PowerPlex 21 kit in degraded samples, 77.56%of 15 autosomal loci were successfully typed by Identifiler Plus kit in degraded samples. Conclusions The highest detection rate was achieved using Identifiler Plus kit, when more loci are required, GlobalFiler kit is more suitable than PowcrPlex 21 kit in typing aged buccal swabs.
