摘  要:目的评价Sysmex UF-1000i尿有形成分分析仪定量计数尿液白细胞和红细胞的性能。方法收集182例尿液标本,对UF-1000i的线性、稳定性、精密度、准确度及携带污染率等指标进行评价,并与人工镜检及AX4280干化学仪器检测的一致性进行比较。同时测定440例健康成人的尿液白细胞和红细胞,用于参考范围的调查。结果该仪器计数白细胞(0~109. 9)/μl和红细胞(0~108. 5)/μl线性良好(r值分别为0. 999 7、0. 997 3),批内和批间精密度的变异系数低于5. 6%,白细胞和红细胞的携带污染率分别为1. 44%、2. 22%,在室温及冷藏的稳定性表明放置4 h和24 h对检测尿液白细胞和红细胞结果无影响。UF-1000i和人工镜检测定尿液白细胞和红细胞结果差异无统计学意义(P> 0. 05),与AX4280仪器一致性好(Kappa值分别为0. 606和0. 598)。男性和女性参考区间分别为白细胞(0~10. 4)/μl、(0~15. 2)/μl,红细胞(0~12. 0)/μl、(0~17. 8)/μl。结论 UF-1000i尿沉渣仪器具有高度精密性,且提高了尿液沉渣的自动化分析能力,可做为常规尿液筛查方法在临床实验室开展应用。Objective To evaluate the performance of counting the urinary WBCs and RBCs that utilize the Sysmex UF-1000 i automated urine analyzer. Methods A total of 182 specimens were collected and we assessed the Sysmex UF-1000 i by analyzing the linear range,stability,reproducibility,accuracy and carryover rate,and then compared with the microscope urine dreg by quantitative determination method. And the consistence was compared with manual microscopy and AX4280 dry chemical instrumentation. At the same time,urine leukocytes and red blood cells of 440 healthy adults were measured for reference range investigation. Results The instrument shows a good linearity(r = 0. 999 7,r = 0. 997 3) in counting the RBCs(0-109. 9)/μl and WBCs(0-108. 5)/μl,in within-run and between-run precision assays,the coefficients of variation for UF-1000 i were less than 5. 6%,the carryover rate was 1. 44% in WBCs and 2. 22% in RBCs,and we respectively placed the specimens in room and refrigerate,which indicated that it made no difference whatsoever to their results in 4 or 24 hours. Results between UF-1000 i and the microscope urine dreg quantitative determination method showed no significant difference(P〈0. 05),and the results were consistent with those of AX4280(Kappa = 0. 606,Kappa = 0. 598). The normal reference range for males and females was WBC:(0-10. 4)/μl,(0-15. 2)/μl,RBC:(0-12. 0)/μl,(0-17. 8)/μl,respectively. Conclusion The UF-1000 i urinary sedimentation instrument is highly precise and improves the automated analysis of urine sediments. It can be used as a routine urine screening method in clinical laboratories.
