摘  要:目的 观察脂多糖 (LPS)诱导D -半乳糖 (D -gal)致衰老大鼠的急性肺损伤 (ALI)及氨基胍 (AG)对其是否有保护作用。方法 大鼠 2 4只随机分成两部分 :6只为正常对照组 ,18只经腹腔注射D -gal复制衰老动物模型。后者再随机分成 3组 :衰老对照组 (6只 )、LPS组 (6只 ,静脉注射LPS 5mg/kg,诱导形成ALI)、AG +LPS组 (6只 ,注射LPS前 30min静脉注射AG 2 5mg/kg)。注射LPS或生理盐水后 2h收集标本待测。结果 D -gal致衰老大鼠较正常对照组大鼠血中乳酸脱氢酶(LDH)活性显著升高 ,而肺组织中Na+ -K+ -ATPase活性下降 (均P <0 0 5 )。衰老大鼠注射LPS后 2h已形成ALI。肺泡灌洗液 (BALF)中蛋白含量增加 (P <0 0 1) ;BAIF中蛋白含量与血浆中蛋白含量比值 (PPI)升高 (P <0 0 1) ;肺湿干重量比增加(P <0 0 1)。血中乳酸 (LD)、丙二醛 (MDA)、一氧化氮 (NO)含量和乳酸脱氢酶 (LDH)活性 ,以及肺组织中髓过氧化物酶 (MPO)活性均显著升高 (均P <0 0 1) ;而肺组织中Na+ -K+ -ATPase活性显著下降 (P <0 0 5 )。预先给予AG可显著地缓解上述指标的变化 (P <0 0 5 ,P <0 0 1)。结论 D -gal致衰老大鼠机体已发生一定的退化性改变。静脉注射LPS可引起衰老大鼠明显的ALI。AG对LPS诱导衰老大鼠的ALI有显著的保护作用。Objective To investigate acute lung injury (ALI) induced by lipopolysaccharide (LPS)in aging rats and the effect of aminoguanidine (AG).Methods 24 Wistar male rats were randomly divided into two parts.6 rats were normal control 18 rats were used for reproducing aging animal model (D-gal 50 mg/kg body weight was injected intraperitoneally, once a day, total 6 weeks). Aging rats were randomly divided into 3 groups: aging control group (6 rats), aging LPS group (LPS 5 mg/kg body weight, intravenous injection,6 rats);AG+LPS(group AG was given 30 min before LPS administration,25 mg/kg,intravenous injection,6 rats).The samples were collected at 2 hours after LPS or saline administration.Results Compared with normal control,in aging rats LDH activity in blood increased (P<0 05)and Na +-K +-ATPase activity in lung tissue decreased(P<0 05).There was obviously ALI in aging LPS rats at 2 hours after LPS administration.Protein content in bronchial alveolar lavage fluid (BALF) and the ratio of protein content in BALF and plasma were increased obviously (all P<0 01).The ratio of lung wet weight and dry weight was increased (P<0 01).LD?MDA?NO - 2/NO - 3contents and LDH activity in blood ,as well as MPO activity in lung tissue all increased significantly (all P<0 01) But,Na +-K +-ATPase activity in lung tissue decreased markedly (P<0 05).Above changes were obviously attenuated in GBE+LPS rats (P<0 05,P<0 01).Conclusions There were some degenerative changes of organic function in aging rats.Acute lung injury was obviously formed by intravenous injection LPS in D-gal induced aging rats.There was markedly protective effect of AG on ALI induced by LPS in aging rats.
