摘  要:本文采用一种生物可降解螯合剂亚氨基二琥珀酸(IDS),制备出一种新型的氨羧类螯合配体色谱填料。通过电位滴定的方法,确定出IDS在硅胶基质上的键合量,筛选出最佳键合配比。用制得的IDS-硅胶柱按照建立的色谱条件分离了蛋白质混合物,考察了IDS-硅胶柱的阳离子交换特性。采用前沿色谱法将金属离子动态固定在IDS-硅胶柱上,利用电感耦合等离子体发射光谱法测定了金属离子在IDS-硅胶柱上的键合量,论证了色谱柱的金属螯合特性。实验结果表明,IDS-硅胶填料既有良好的阳离子交换特性,可用于分离纯化带电荷的生物大分子,还具有优良的金属螯合特性,可用于工业废水中重金属离子的去除。A novel chromatographic packing with an aminocarboxyl chelating ligand was prepared with iminodisuccinic acid(IDS),a biodegradable chelator.The bonding amount of IDS was obtained by potential titration method and the optimal ratio of IDS/silica was selected.According to the established chromatographic conditions,cation exchange characteristics of IDS-Silica column were examined by separating protein mixture with the obtained column.Metal ions were dynamically immobilized on IDSSilica columns by frontal chromatography.Metal chelating properties of IDS-Silica column were proved by determining the bonding amounts of metal ions on the columns with inductively coupled plasmaoptical emission spectrometry(ICP-OES).The experimental results showed that IDS-Silica packing exhibited a good cation exchange behavior for separating and purifying bio-macromolecules with charge,and also displayed an excellent metal chelating property for removing heavy metal ions from industrial wastewater.
