摘 要:建立微乳液胶束电动色谱法(MEEKC)同时测定白屈菜中6种异喹啉类生物碱成分。实验以20mmol/L十二烷基硫酸钠,50mmol/L脱氧胆酸钠,0.3%(V/V)乙酸乙酯,8%(V/V)正丁醇,91.7%(V/V)的5 mmol/L硼砂-10 mmol/L磷酸溶液(pH=4.0)为运行缓冲液,25kV为分离电压。以214nm和280nm为检测波长。在上述分析条件下,6种分析物在20min内达到基线分离。经方法学验证,该MEEKC法线性关系良好,精密度高,应用于白屈菜全草提取物中异喹啉类生物碱的测定,回收率良好,可以为白屈菜中异喹啉类生物碱的测定提供一种新方法。A microemulsion electrokinetic chromatrography(MEEKC) method was established to determinate six isoquinoline alkaloids in Chelidonii herba.Experiments were carried out with 20 mmol/L SDS,50 mmol/L SDC,0.3%(V/V)ethyl acetate,8%(V/V)n-butanol,and 91.7%(V/V)5 mmol/L borax-10 mmol/L phosphoric acid solution(pH=4.0)for the running buffer,214 nm and 280 nm for the detection wavelength,and 25 kV for the separation voltage.Under the above conditions,the six analytes were baseline separated within 20 min.After methodology validation,the MEEKC method showed a good linear relationship and high precision.The established method was successfully applied to the determination of alkaloids in Chelidonii herba and the recovery was satisfactory,indicating that the MEEKC could provide a new method for the determination of isoquinoline alkaloids in Chelidonii herba.
