摘 要:目的:观察当归对自发性高血压大鼠血管内皮组织miR122靶基因SLC7A1表达的影响,探讨其影响高血压发生的作用机制。方法:将自发性高血压大鼠随机分为模型组、当归挥发油组(低、中、高剂量组)、缬沙坦组,同周龄Wistar大鼠为正常对照组,每组8只,模型组及正常对照组给予等量生理盐水灌胃。QT-PCR法检测SLC7A1的mRNA表达水平,Western Blot法检测各组大鼠血管内皮组织SLC7A1蛋白表达量。结果:与模型组比较,当归挥发油高、中、低剂量组及缬沙坦组SLC7A1的mRNA表达水平均显著下调(P〈0.05);当归挥发油各组中SLC7A1的蛋白表达水平亦显著下调(P〈0.05)。结论:当归通过miR122下调SLC7A1的表达对血管内皮发挥作用。Objective:To study the effects of volatile oil from Radix Angelicae Sinensis on SLC7A1 gene expression of mi R122 in spontaneously hypertensive rats(SHR),and to explore the mechanism of its effect on hypertension. Methods:40 SHR rats with spontaneously hypertensivewere divided into Radix Angelicae Sinensis group, model control group,valsartan group randomly,Wistar rats of the same age as normal control group.The model group and the normal control group were treated with the same volume of normal saline by intragastricad ministration.The m RNA levels of SLC7A1 was detected by QT-PCR and the protein expressions of SLC7A1 was tested by Western Blot.Results :Compared with the model group,the mRNA and protein expressions of SLC7A1 were significant decrease in Radix Angelicae Sinensis group and valsartan group(P〈0.05).Conclusion:Radix Angelicae Sinensis could interfere the blood pressure by acting on miR122 through down regulation the expression of SLC7A1 gene.
