摘  要:目的:观察消渴通痹颗粒含药血清对高糖环境下EPCs PI3K/Akt/e NOS分子信号通路的影响。方法:体外培养并鉴定大鼠骨髓EPCs,制备消渴通痹颗粒含药血清,对EPCs进行不同高糖血清或PI3K抑制剂Wortmannin及e NOS抑制剂L-NAME的干预,检测EPCs的增殖及小管形成能力,Western-blot检测Akt、p-Akt及e NOS、p-e NOS蛋白表达。结果:与对照组比较,消渴通痹颗粒含药血清能促进EPCs的增殖及小管形成,不同浓度的消渴通痹颗粒含药血清预处理后在一定程度上阻止了AKT,p-AKT,e NOS,p-e NOS水平的下降(P<0. 05)。结论:消渴通痹颗粒可能通过上调PI3K/Akt/e NOS分子信号通路相关蛋白的表达水平,促进高糖环境下EPCs增值和小管形成。Objective: To observe effect of drug serum containing"Thirst Dissipating and Numbness Removing"Granule on EPCs PI3 K/Akt/e NOS molecular signaling pathway in high glucose environment. Methods: Culturing and identifying EPCs of rat bone marrow in vitro,preparing drug serum containing"Thirst Dissipating and Numbness Removing"Granule,intervening EPCs with different high glucose serum or PI3 K inhibitor Wortmannin and e NOS inhibitor L-NAME,testing the proliferation and tubular formation ability of EPCs,detecting Akt,p-Akt and e NOS by Western-blot,and the expression of p-enos protein. Results: Compared with the controlled group,the drug serum containing "Thirst Dissipating and Numbness Removing"Granule can promote the proliferation and tubular formation of EPCs,different concentrations of drug serum containing"Thirst Dissipating and Numbness Removing"Granule had prevented the decline of AKT,p-AKT,e NOS,p-e NOS levels to some extent( P〈0. 05). Conclusion: "Thirst Dissipating and Numbness Removing"Granule may up-regulate the expression level of related protein of pi3 k/akt/enos signaling pathway,to promote EPCs increment and tubule formation in the high glucose environment.
